Objective:To explore the effect of caffeine on moxibustion treat ulcerative colitis in Kunming mice and the expression level of adenosine A1 receptor in hippocampus.Methods:Kunming male mice with weight of 32g±2g were selected as the research object.The ulcerative colitis model was established by the Dextran Sulfate Sodium(DSS)method with 3% dextran sulfate.After 5 days,the UC model of mice were sucessfully established,8 healthy mice were selected as the control group,the other mice were randomly divided into model group,model+moxibustion group,moxibustion +model+ 0 n M caffeine,model+ moxibustion+0.05 n M caffeine group,model+moxibustion +0.15 n M caffeine group,model + moxibustion +0.25 n M caffeine group,8 mice in each group.Moxibustion group chosed Guanyuan point with moxibustion treatment daily for five days,15 min each time,moxibustion +different concentrations of caffeine group were administered via gavage daily for five days before moxibustion,the control group and the model group were only bundled without treatment.Observe the general condition and the score of disease activity index(DAI);24 hours after the end of treatment,we got the hippocampus,using real-time PCR to detect the expression level of A1 R m RNA.Statistical analysis use SPSS23.0.Results:1.Study on the treatment effects of moxibustion in the UC miceAfter establishment of the models,the mice DAI score notably increases.After the moxibustion treatments,the DAI score decreases.Comparison between the treatment group and the model group shows that the DAI score is statistically significant(P <0.05),and no obvious difference exists between the two groups(P > 0.05).2.Study on the treatment of coffeine in the UC miceAfter caffeine treatment,the mice in the model + 0.05 nM caffeine group,model+0.15 n M ca 15 n M and modle+0.25 n M.3.Effect of Moxibustion Combined with Caffeine on UC MiceCompared with the moxibustion moxibustion group,there was no significant difference in the DAI score between the moxibustion+0.05 n M caffeine group and the moxibustion+0.15 n M caffeine group(P>0.05);moxibustion+0.25 n M caffeine group mice and moxibustion moxibustion group Compared with DAI score,there was a difference between groups(P<0.05)4.Changes of A1 R m RNA Expression in hippocampus.Mode group was compared with the control group,the expression of A1 R m RNA in the hippocampus was up-regulated 2.4-fold;Model+moxibustion group was compared with the model group,the expression of A1 R m RNA in the hippocampus was down-regulated by 4.9-fold.Compared with the model group,A1 R m RNA expression in the hippocampus of model + 0 caffeine group,model + 0.05 n M caffeine group,model + 0.15 n M caffeine group,model +0.25 n M caffeine group were up-regulated0.2-fold,1.6-fold,2.4-fold,3.1-fold,respectively;Compared with model group moxibustion + 0n M caffeine group,moxibustion +0.05 n M caffeine group moxibustion + 0.15 n M caffeine group,moxibustion +0.25 n M caffeine,the expression of hippocampus’ A1 R m RNA was down-regulated by4.45-fold,3.8-fold,2.2-fold,1.9-fold,respectively.Conclusions:1.Moxibustion has a good therapeutic effect on DSS-induced UC model mice;2.Caffeine gavage has anti-inflammatory effects on UC mice,and 0.05 n M,0.15 n M,0.25 n M caffeine have similar therapeutic effects.After 0.25 n M caffeine was given,the effect of moxibustion on UC has been inhibited.3.Adenosine A1 receptor in hippocampus may participate in moxibustion’s treatment of UC mice and be involved in caffeine’s inhibition of UC treatment.
