Every Ginger Moxibustion For Treatment Of Ulcerative Colitis, Liver Depression And Spleen Experiments

1 ObjectiveUlcerative colitis(UC), that is chronic nonspecific ulcerative colitis, is an chronic and recurred disease characterized by chronic inflammation and ulceration of colonic mucous membrane. Clinical manifestation: abdominal pain, diarrhea, bloody purulent stool, mucous stool, tenesmus, and so on. Since it's a disease which is difficult to heal and it has a large chance to recur again, WHO has defined it as a disease which is difficult to cure. And it also became a hotspot in the field of Gastroenterology.In 1956, the first patient with UC in our country was reported. With the improvement of living standard and lifestyle, the incidence of this disease has a adscendent tendency. The etiopathogenisis is not elucidated well, and it is considered that UC has something with many factors, such as genetic factor, affectoi, psychologic factor, immunization factor, et al. Recently, the researches concentrated the immune disfunction. The main treatment prescription includes maintenance therapy and the improvement of symptom. However, these can just relieve the pathogenetic condition and can't cure the disease.Because the patients suffer UC for a long time, they feel like a new kind of treatment which is painless, convenient and eutherapeutic. Ginger-partition moxibustion is a traditional treatment of TCM. It has some fortes, for example, the patients can treat themselves at home because of the convenient use. In addition, it is cheaper, painless and acceptive. Many doctors had treat the patients by using ginger-partition moxibustion and get good therapeutic effect. But, there are few studies about the therapic mechanism of ginger-partition moxibustion. So, we design this experimental reseach.We choose the TCM syndrome of"Stagnation of liver-qi and Deficiency of spleen-qi"as the object, observe the therapeutical effect of ginger-partition moxibustion to UC and the influence of ginger-partition moxibustion to the cytokine and neuropeptide. Thus, we want to investigate the internal mechanism.2 MethodsThe experimental reseach includes three rests. Rat model of ulcerative colitis with the TCM syndrome of"Stagnation of liver-qi and Deficiency of spleen-qi"is induced by trinitro-benzene-sulfonic acid (TNBS)/ethanol solution and constraint.Test one observed the influence of gingers-partition moxibustion and SGJP on rats of model group. Observe the general state of health of the rats after making model and give a mark of DAI. After treating 3 weeks, get the colon from anus to ileocecal junction and observe mucous and give a mark of CMDI.Test two investigated the influence of gingers-partition moxibustion and SGJP on the content of VIP. After treating 3 weeks, get the distal end of colon that is about 8cm long and determine the content of VIP.Test three studied the influence of gingers-partition moxibustion and SGJP on the content of IL-1βand IL-10. After treating 3 weeks, get the distal end of colon that is about 8cm long and determine the content of IL-1βand IL-10.3 Result3.1 General state of health: The stools of the rats of model group is loose stools after model-making. With the lasting of time, there are some symptoms, such as mucous stool, bloody purulent stool, piloerection, emaciation, extrado, gathering, lazy, little food and water, loosing weight. The rats'general conditions of all treatment groups after 3 weeks'treatment are better than that of model group.3.2 Pathomorphological changes of colonic tissue: There are acute and chronic changes and ulcers on the colons of the rats of model group. After 3 weeks, all ulcers vanish, and there aren't inflammatory cells in the colonic tussue.3.3 The disease activity index(DAI): DAI increases after model-making and decreases after treating.3.4 The colonic mucous membrane damage index(CMDI): CMDI of the treating group decreases more than that of the model group.3.5 Content of VIP: VIP content of model group show a distinct increase and is significant (P<0.01) compared to normal group. VIP content of treating group show a distinct decrease and is significant (P<0.01) compared to model group.3.6 Content of IL-1β: The content of IL-1βof model group has an obvious increase than that of normal group(P<0.01). The content of IL-1βof all treating group has an obvious decrease than that of normal group(P<0.01).3.7 Content of IL-10:The content of IL-10 of model group has an obvious decrease than that of normal group(P<0.01). The content of IL-1βof all treating group has an obvious increase than that of normal group(P<0.01).4 ConclusionRat model of ulcerative colitis with the TCM syndrome of"Stagnation of liver-qi and Deficiency of spleen-qi"is induced successfully by TNBS/ethanol solution and constraint.Gingers-partition moxibustion can reduce the expression of IL-1βand VIP and raise the expression of IL-10. It may treat UC by adjusting neuropeptides and cytokines.
