Objective:Taking psoriasis patients with blood-heat syndrome of psoriasis as the research object and using healthy people as control,the clinical curative effect of psoriasis vulgaris(blood heat syndrome)treated by Jiawei Liangxue Xiaofeng Powder was evaluated by PASI score;Western Blot technique was used JAK/STAT signal transduction pathways were examined to summarize the characteristics of psoriasis blood-heat syndrome based on JAK/STAT signal transduction pathway cell apoptosis,which is psoriasis vulgaris The evidence provides a microscopic material basis;At the same time,through comparison of experimental data before and after treatment of Jiawei Liangxue Xiaofeng Powder,the effect of Jiawei Liangxue Xiaofeng Powder on apoptosis and its mechanism are explored,providing new ideas for the treatment of psoriasis vulgaris.Methods:Ten psoriasis patients with blood-heat syndromes and healthy subjects who met the inclusion criteria were taken as the experimental group and the control group,respectively.The experimental group was treated with self-control before and after treatment,and the change of skin lesions before and after the administration of Jiawei Liangxue Xiaofeng Powder was observed and the PASI score was performed.Western Blot technique was used to detect the apoptosis-related factors bcl-2,p53 and key proteins JAK3,STAT3 of the JAK/STAT signal transduction pathway before and after treatment in the experimental group,and the data were analyzed statistically.Result: 1.After comparing PASI values before and after treatment of Jiawei Liangxue Xiaofeng Powder,it was found that the PASI score of the experimental group was significantly lower than that before the treatment(P<0.05).2.Compared with the control group,the expression of bcl-2,JAK3 and STAT3 genes in the experimental group was increased,and the difference was significant (P<0.05).However,the expression level of p53 in the experimental group was lower than that in the control group,and the difference was statistically significant.(P<0.05).3.In the comparison of the experimental group before and after treatment,the expression of bcl-2,JAK3,and STAT3 in the serum of patients treated with Jiawei Liangxue Xiaofeng Powder was lower than that before treatment,and the difference was statistically significant(P<0.05).The expression level of p53 was higher than before treatment,and the difference was significant(P<0.05).Conclusion: 1.After treatment with Modified Jiawei Liangxue Xiaofeng Powder,the PASI score of patients with psoriasis blood-heat syndrome was significantly lower than that before treatment.Jiawei Liangxue Xiaofeng Powder can reduce the skin lesions of psoriasis blood-heat syndrome.2.In this study,the expression levels of JAK3,STAT3 and bcl-2,the key proteins of JAK/STAT signaling pathway in peripheral blood of psoriasis vulgaris patients with psoriasis vulgaris were highly expressed compared with those in healthy。people,The low expression of pro-apoptotic gene p53 can confirm that JAK/STAT pathway and apoptosis are involved in the development of psoriasis blood-heat syndrome,high expression of JAK3,STAT3,bcl-2 and low expression of p53.Is part of the microscopic material basis of psoriasis blood heat syndrome.3.Jiawei Liangxue Xiaofeng Powder can down-regulate JAK3 and STAT3 levels in the serum of patients with psoriasis blood-heat syndrome and inhibit the activity of JAK/STAT signaling pathway;at the same time,it can down-regulate bcl-2 and up-regulate the expression of pro-apoptosis gene p53.Level induces apoptosis.4.The inactivation of JAK/STAT signaling pathway may be an important mechanism of Jiawei Liangxue Xiaofeng Powder in inducing cell apoptosis.Jiawei Liangxue Xiaofeng Powder can induce apoptosis by inhibiting the activity of JAK/STAT signaling pathway and achieve therapeutical vulgaris.The purpose of psoriasis blood heat syndrome. |
