Objective: The 5-HT signal pathway in the brain-intestinal axis was used as the starting point to analyze the relationship between the 5-HT signal pathway and the visceral hypersensitivity of irritable bowel syndrome diarrhea-predominant(IBS-D).To study relieving the mechanism of visceral hypersensitivity in irritable bowel syndrome diarrhea-predominant(IBS-D)with Tong Xie Yso Fang of the "suppression of wood" group-the Bai Shao Fang Feng components.method: 1、90 cases of IBS-D patients who met the inclusion criteria and signed the informed consent were randomly divided into the Bai Shao Fang Feng group,the Chen Pi Bai Zhu group and the whole group.Each patients by rule to taking the corresponding components drugs for four weeks.2、To observe and record each IBS-D patients of the monthly normal defecation maintenance time and abdominal pain / abdominal discomfort and anxiety and depression self-rating scale score,at the time of into the group and after treatment.3、Blood samples were collected from each IBS-D patient at the time of into the group and after treatment,serum and plasma were extracted after centrifugation.And whole blood samples were extracted with PA Xgene TM whole blood RNA tube.Finally,the plasma was detected by double-antibody sandwich ABC-ELISA method.The serum and whole blood samples were gene chip detected by PCR gene detection technology.4、Statistics each IBS-D patients of the monthly normal defecation maintenance time and abdominal pain / abdominal discomfort and anxiety and depression self-rating scale score.The expression of 5-HT signaling pathway(SLC6A4,HTR3 A,HTR4,HTR7)was extracted from gene detection of the original data.The data of 5-HT in plasma biomolecule detection were extracted.Use SPSS 21.0 statistical software for statistical analysis of the data,Show was statistically significant to P<0.05.Result:1、There were no significant differences in the baseline,age,course of disease between all groups before treatment(P> 0.05).2、Each group of IBS-D patients had abdominal pain / abdominal discomfort,the monthly normal defecation to maintain a shorter time,mild or moderate anxiety and depression.After treatment,each group with abdominal pain / abdominal discomfort,anxiety and depression were significantly alleviated,monthly normal defecation time increased significantly(P <0.05).Among them,the Bai Shao Fang Feng group and the whole group of symptoms relief significantly,and between the Chen Pi Bai Zhu group was significant difference(P <0.05).2、There was no significant difference in plasma 5-HT level between the two groups(P> 0.05).After treatment,plasma 5-HT levels in the Bai Shao Fang Feng group and the whole group of patients with IBS-D were significantly lower than those before treatment(P <0.05).But the Chen Pi Bai Zhu group was no significant difference(P> 0.05).4 、 The expression of SLC6A4 m RNA in 5-HT signaling pathway was significantly lower than that in normal group(P <0.05),The expression of HTR3 Am RNA was significantly higher than that of normal group(P <0.05),The expression of HTR4 m RNA and HTR7 m RNA was not significantly different from that of normal group(P> 0.05).After treatment,the expression of SLC6A4 m RNA was significantly up-regulated in the Bai Shao Fang Feng group and the whole group(P <0.05),and the expression of HTR3 Am RNA was significantly down-regulated(P <0.05).But the expression of SLC6A4 m RNA and the expression of HTR3 A m RNA were not significantly decreased in the Chen Pi Bai Zhu group(P> 0.05).Conclusion: 1 、 Tong Xie Yao Fang be able to effectively alleviate IBS-D patients with abdominal pain and diarrhea symptoms and anxiety and depression,and Bai Shao Fang Feng is to alleviate IBS-D symptoms of the main components;2、The 5-HT signaling system in the brain-intestinal axis may be involved in the formation of IBS-D visceral hypersensitivity;3、Tong Xie Yao Fang of the Bai Shao Fang Feng components may be by adjusting the 5-HT signaling pathway to alleviate the visceral hypersensitivity state of IBS-D,thus alleviating the clinical symptoms of IBS-D.
