Objective:In this study,we compared the expression of GRP78 and CHOP protein in hyperlipidemic pancreatitis(HLP)rats,and discussed the role of endoplasmic reticulum stress(ERS)related molecules in the process of HLP progression and aggravating the progression of HLP.Methods:40 SD rats were randomly divided into 4 groups: control group(Group sham),pancreatitis group(group AP),hyperlipidemia group(group HTG),hyperlipidemic pancreatitis group(HLP group),10 rats in each group.The rats in group Sham and group AP were fed with normal diet,and the high fat diet of group HTG and HLP group was fed for 8 weeks.The high fat model was constructed and the operation was prepared.All rats were fasted 12 h before operation and were free of drinking water.Group AP and group HLP were treated with L-arginine(PH=7200 mg / 100 g,interval 1 h,divided into2 times)by intraperitoneal injection,and then the SD model of pancreatitis was established.Sham group and HTG group were converted to normal saline by injection.After 6 h of operation,the serum amylase content was detected.HE staining was used to observe the histopathological changes of the pancreas in rats.The expression of GRP78 and CHOP was detected by Western blot.Results:1.After 8 weeks,the level of TG in the high fat group was 1 times higher than that in the normal diet group,and the high fat model was established successfully.2.The level of serum amylase in group AP and group HLP was significantly higher than that in group Sham,and the model of 6h was successful.3.Histopathology results showed that compared with Sham group,HLP group and AP group had obvious edema compared with Sham group,with varying degrees of bleeding and some cell necrosis,and the degree of lesion in HLP group was heavier than that in AP group,and the pathological injury score was higher than that in AP group.4.The results of Western blot showed that GRP78 and CHOP were expressed in both group AP and HLP group,and the expression level of HLP group was higher.Conclusion:Endoplasmic reticulum stress is not only involved in the pathogenesis of hyperlipidemia related pancreatitis,but also induces the apoptosis of pancreatic cells and aggravates the development of hyperlipidemic pancreatitis.
