Object:To evaluate the therapeutic effect and mechanisms of Essential Oil of Ligusticum chuanxiong Hort on vascular dementia,and evaluate its protective.Methods:?1?the acute toxicity of LEO on mice in accordance with classical acute toxicity test methods.?2?Chronic hypoperfusion vascular dementia rats model induced by ligation of bilateral common carotid artery,a month after the surgical proce-dure,rats would be intragastric administration of LEO?7.7mg/kg,23mg/kg,77mg/kg?for consecutive eight weeks.Their spatial learning and memory abilit-ies were assessed using the Morris water maze.The level of acetylcholine?Ach?,acetylcholinesterase?Ach E?,Superoxide dismutase?SOD?,malondialdehyde?MDA?,nitric oxide?NO?,nitric oxide synthesis?NOS?,Monoamine oxidase?MAO?in cerebral cortex tissue and HDL-C,LDL-C,TG,TC in serum were detected.The content of norepinephrine?NE?,dopamine?DA?and 5-hydroxytrypt-amine?5-HT?in cerebral hippocampus tissue were detected by fluorometry.The rat blood rheology parameters were detected by blood rheometer.Pathological changes of cerebral cortex were observed.?3?Oxidative damage cell model induced by H2O2 were used to evaluate the effect of antioxidant of LEO;?4?Memory disorders induced by intraperiton-eal injection of 2 mg/kg scopolamine,or intragastric administration of 0.01 ml/g 40%ethanol were used to observe the effect of LEO in mice.Results:?1?The LEO could cause the mice to be poisoned,the LD500 of LEO is1.167 g·kg-1·d-1(the 95%confidence interval is 1.03551.3304 g·kg-1·d-1).?2?Compare with the sham group,the average escape latency was significantly longer in model group,the distance swimming of VD model rats were increased.during probe trials,the time of first across the platform of VD model rats were significantly more than sham-operated rats,the number of swimming across the platform within 90s was reduced,and percentage of the second quadrant residence time/total time?%?,platform residence time/total time?%?was reduced in morris water maze test.LEO can significantly shorten the average escape latency,reduce the distance swimming of rats.during probe trials,LEO could reduce the time of first across the platform,increase the number of swimming across the platform within 90s,and increase percentage of the second quadrant residence time/total time?%?,platform residence time/total time?%?in morris water maze test.LEO could increase the levels of Ach and SOD,reduce the levels of AChE,MDA,NO and iNOS,TNOS and MAO in cerebral cortex of rats.Increase content of NE,DA and 5–HT in the hippocampus of rats,it could increase HDL-C,reduce the TG in serum.improved the whole blood viscosity and plasma viscosity.The neurons of the sham group were clear,the staining were uniform,the distribution were neat and the number was more.In contrast,The model group due to cell damage,nuclear condensation,the number of stained cells decreased.Compared with the model,the number of cells in LEO medium and high dose group increased correspondingly.?3?LEO could increase PC12 cell survival rate injuried by H2O2;LEO 0.1?g/L,1?g/L,5?g/L could significiantly reduce the level of LDH in H2O2 PC12.?4?compared with the sham-operated,treated with scopolamine or ethanol shortened the step-down latency and increased the step-down errror times significantly in the step-down passive avoidance test whin 5minutes in mice.LEO could prolong the step-down latency significantly.Conclusion:The LEO could cause the mice to be poisoned,and could significantly improve the learning and memory disorders;Have obviously therapeutic effect on VD,The mechanism may be related to reduce AChE activity and increase the level of Ach in the cerebral cortex,Reduce the peroxidation damage,modulated the hippocampus neuro transmitter,improve blood rheology and blood lipids.
