Effect Of Danshen Injection On TGF-β1,TNF-α And IL-6 In Serum Of Rat Model With Chronic Pancreatitis

Objective:To observe the effect of Salvia miltiorrhiza injection on the level of TGF-β1,TNF-α and IL-6 in the serum of rats with chronic pancreatitis(CP).Methods:30 male SD rats were randomly divided into Normal group、Modal group and Intervention group after 1 week of normal feeding.Normal group rats were received an intramuscular injection of the same dose of saline daily;Model group and Intervention group rats were intravenous injected with DBTC by 8mg/kg to make animal model.24 hours later,the model group rats were injected with the same dose of saline and the Intervention group was received a daily intramuscular injection of Danshen injection by1600mg/kg.After 4 weeks,all rats were killed and blood samples and tissue samples were collected.The levels of TGF-β1,TNF-α and IL-6 in blood were measured by ELISA method,and pancreatic tissue samples were taken for pathological examination.Results:In 30 animals,7 died and the rat of survived 23,and 12 successful models were confirmed by histopathological observation of the pancreas.The histopathological score of rats in model group was the highest,while normal group was the lowest and the differences between groups were statistically significant(P < 0.05).The cytokines blood leve of TGF-β1,TNF-α and IL-6 of model group were(65.08±5.31)pg/ml,(188.88±22.17)ng/ml,(110.43±16.11)ng/L,significantly higher than those of normalgroup(P < 0.05),the Intervention group compared with the model group that TGF-β1,TNF-α and IL-6 levels were(55.55±4.40)pg/ml,(148.26±18.43)ng/ml,(87.92±12.24)ng/L,lower than those in the model group,not to normal,but still higher than normal and the differences were statistically significant(P < 0.05).Conclusion:The drug of Danshen can inhibit the secretion of certain cytokines in chronic pancreatitis rat model,and the findings from pathological changes shows that pancreatic fibrosis were alleviated under the situation of treatment of Danshen injection.The mechanism of anti fibrosis of the pancreas may be inhibiting the expression of TGF-β1,TNF-α and IL-6 to reduce pancreatic fibrosis.
