Hui-hui Gan-song Yin Improve Zuker Diabetic Fat Mouse Early Metaphase Renal Fibrosis MiR-21 Related Molecular Mechanism Research

Objective:To verify Hui-hui Gan-Song Yin（HGY）on diabetic ZDF protection of early metaphase renal fibrosis,reveal HGY is given priority to with miR-21 targets,PTEN/Akt signal transduction pathway,to improve renal function and relieve renal fibrosis.Methods:Using the diabetes ZDF successful build DM rats model of renal fibrosis（6weeks ZDF rat feeding 8 weeks）,and then were given HGY and positive control drug Fosinopril Sodium Tablets（Fos）after the intervention,to observe the general situation of rats,kidney index,blood sugar,and by using light microscopy and transmission electron microscope kidney tissue morphology and pathological changes.Using Western-Blot technique,observation of FN,ColⅠ,TGF-beta1,PTEN,PKB,Akt protein expression in rat kidneytissues;WithRT-PCRtechniquetoobservemiR-21 andFN,ColⅠ,TGF-beta1,PTEN,PKB,Akt mRNA expression in rat kidney.Results:1.Significant ZDF rats polydipsia,polyphagia,polyuria,emaciation,depressed,hair dry peeling performance,water quantity and obviously increase food intake;HGY improve ZDF rats fasting plasma glucose（after 20 weeks）,left and right kidney weight（P<0.05）;Histopathological（HE staining and Masson,three color method,transmission electron microscope,immunohistochemical method）the results showed that HGY told ZDF rat early metaphase DKD glomerular,renal tubule and interstitial have obvious repair and protection.2.The RT-PCR method detection results show that compared with ZL group,ZDF miR-21,FN,ColⅠ,TGF-beta1,Akt mRNA level increased significantly（P<0.01）,PTEN mRNA level significantly decreased（P<0.01）;Compared with ZDF group,ZDF+Fos,ZDF+HGY10g·kg^-1,ZDF+HGY5g·kg^-1group,miR-21,FN,ColⅠ,TGF-beta1,Akt mRNA level increased significantly（P<0.05or0.01）,ZDF+Fos ZDF+HGY10g·kg^-1group,ZDF+HGY5gkg^-1set of PTEN mRNA level decreased significantly（P<0.05or0.05）.The Western Blot method detection results show that compared with Compared with ZL group,ZDF FN,ColⅠ,TGF-beta1,Akt protein expression level increased significantly（P<0.01）,the expression levels of PTEN protein significantly declined（P<0.01）;Compared with ZDF group,ZDF+Fos ZDF+HGY10g·kg^-1group and ZDF+HGY5g·kg^-11 group FN,ColⅠ,TGF-beta1,Akt protein expression level was significantly increased（P<0.05or0.01）,ZDF+Fos ZDF+HGY10g·kg^-1group,ZDF+HGY5g·kg^-1setofPTENproteinexpressionleveldecreased significantly（P<0.05or0.05）.Conclusion:Middle ZDF HGY on diabetes rat renal fibrosis plays a protective role,may give priority to with miR-21 targets,PTEN/Akt signal transduction pathway,middle to improve renal interstitial fibrosis.
