Pouzolzia zeylanica(L.)Benn.Var.microphylla(Wedd.)W.T.Wang(PZ)is one of the variations from Urticaceae of Pouzolzia zeylanica.It is mostly used to treat syphilis,inflammation and trauma in folk with significant effects of corruption pus and lighting inflammation.Previous experiments have established that PZ extract has obvious effect of anti-inflammatory,analgesic,accelerate wound healing and 36 compounds were identified from it.The structure types of compounds are mainly lignans and flavonoid glycosides,but most of the compounds have not been studied for efficacy.In this study,the compounds were isolated and identified by chromatography and spectrometry,and the antibacterial and anti-inflammatory activity of each compound was preliminarily studied.The paper provides scientific basis for elucidation of the material basis and mechanism of detoxification of PZ.The main research contents are asfollows:1.Studies on chemical constituents of ethyl acetate from PZ.The chemical constituents of ethyl acetate were systemmatically studied with silica gel,ODS and Sephadex LH-20 column chromatography,preparation of liquid phase and spectral analysis.17 compounds were obtained.Their structures were identified by spectral characteristics and reported in the literature.They are Pouzolignan K-a(1),Pouzolignan K(2),Pouzolignan F(3),Pouzolignan G(4),Pouzolignan H(5),Pouzolignan I(6),quercetin-3,7-O-?-L-dirhamnoyranoside(7),kaempferol-3,7-O-?-L-dirhamnoyranoside(8),fiedelin(9),(+)-dehydrovomifoliol(10),saropeptide(2-(2-benzamido-3-phenylpropanamido)-3-phenylpropyl acetate(11),3-hydroxy-4-methoxybenzoic acid(12),4-hydroxybenzoic acid(13),protocatechuic acid ethyl ester(14),protocatechualdehyde(15),syringic acid(16),protocatechuic acid(17).Among the 17 compounds,the structural types refer to lignans(1-6),flavonoids(7-8),pentacyclic triterpenoids(9),?,?-unsaturated ketones(10),alkaloids(11)phenolic compounds(12-17).Compound 1 is a new compound,Compound 10-17 were isolated from Pouzolzia genus for the first time.2.Studies on HPLC fingerprint of ethyl acetate: HPLC fingerprint of ethyl acetate in PZ was established by Chinese medicine fingerprint similarity system.12 peaks(compounds 1 to 6,12 to 17)were identified comparison with the obtained compound chromatograms.The ratio of each compound to ethyl acetate was obtained according to the formula.The ratio of compound 1 to 6 is 4:5:1:1:6:2,and the content ratio of simple phenolic compounds 12 to 17 is 3:2:10:6:8:9.3.Studies on the antibacterial activity of compounds.The antimicrobial activity of the compounds was preliminarily tested with the agar disc diffusion and candida albicans,bacillus thuringiensis,staphylococcus aureus,bacillus subtilis,streptococcus pyogenes,escherichia coli and shigella dysentery bacillus as the indicator bacteria.The coordination between the compounds was invenstigated.The dose of compound is 50?g,and the positive control dose is 10?g.The results were as follows: among the fungal susceptibility to Candida albicans,Pozolignan K-a had the strongest activity with moderately sensitive with 50?g,and the inhibition zone reached 13.56 mm.Secondary,compound 6 and Compound 13 has mildly sensitive and the inhibition zone is about 9.00 mm.Compound 17 showed mild susceptibility to bacteria except candida albicans and streptococcus pyogenes,and had extensive antibacterial activity.There was no apparent antibacterial synergy between the various compounds.There was no significant anti-inflammatory activity on the combination of the compounds.4.Studies on anti-inflammatory activity of compounds.The antiinflammatory activity of 17 compounds in human fibroblast cell line HSP was preliminarily tested with inflammatory model induced by LPS.The results showed that: Compounds 1-6,10,13,15 and composition 1 had different degrees of inhibitory effect on the release of IL-6,indicating that these 9 compounds had a certain in vitro anti-inflammatory effect,and the decarbonizing lignan compounds were particularly remarkable.The combined use of descending carbon lignan showed significant anti-inflammatory effects.
