Objective:Based on currently understanding of the toxicity and toxic substances of Araceae herbs,and the current edition of processing method in Chinese Pharmacopoeia,changing the way of improveing and optimizating of the processing technology of Rhizoma Arisaematis,in order to achieve the targets of efficacy enhancing and toxicity reduction,so as to guarantee the quality of the processed products of Rhizoma Arisaematis and enhance the clinical efficacy.At the same time,the study also provide a theoretical basis for the reasonable processing in guiding Araceae medicinal materials.Methods:(1)As the calcium content of calcium oxalate crystal was changed after calcium oxalate,so we use the alcium ion as the final target.Both the single factor analysis and orthogonal test is usded to design the study,with the four main effecters of ethanol concentration,ethanol solution’s pH,infiltration time and temperature,to improve and optimizat the processing technology of traditional Chinese medicine of Araceae processing research;(2)Using the irritation test,acute toxicity and subacute toxicity test,combine with the assaying of the aluminum ion content,to evaluate the safety of the processed products of Rhizoma Arisaematis;(3)Using the experimental method of pharmacology,with the analgesia,sedation and anti-seizure test,to evaluate the effectiveness of the processing products of Rhizoma Arisaematis;(4)According to the Pharmacopoeia method,we assaying the moisture,ash content and water soluble extract,using the UV spectrophotometry to assaying the total flavonoids,to control the quality control of new processed products of Rhizoma Arisaematis.Results:(1)By means of single factor analysis and orthogonal test,the optimal Arisaema new processing technology is to be choosed.The new processing is:Arisaema crude pieces run through with pH4’s 50%ethanol solution,placed in a closed environment under 50 DEG C,moistening 48h.According to this method of processing Rhizoma Arisaematis,most of the calcium oxalate crystal in medicinal materials has been is dissolved,in this way we have achieve the objective of reducing or eliminating the toxicity and stimulation.(2)The assaying of aluminum ion content was tested.The results showed that,the aluminum ion content is significantly lower in the new processed products of Arisaema than was significantly lower than that of the proceessing of products in Pharmacopoeia.It has reduced the aluminum ion effects of the damage to human body;The irritation test results show that,the effect of Arisaema new processed products on rabbit conjunctival irritation is very small,the irritation is lesser than the crude and the Pharmacopoeia method processed products;The acute toxicity test showed that,the LD50 dose of the mixed suspension and water decoction of Arisaema’s new processed products could not be detected.The maximum dose of water decoction equivalent to 2800 times clinical,and the animals toxic reaction is not obvious.We can conclude that products of Arisaema has no apparent toxicity,and it is relatively safe.The subacute toxicity test results showed that,rats for 30 consecutive days intragastric administration to give the water decoction of Rhizoma Arisaematis new processed products,through on the behavior of rats,weight,blood and biochemical indexes and pathological observation,there is no significant impact.We can conclude that said Rhizoma Arisaematis new processed products has low toxicity.(3)The hot plate method and torsion body method of analgesic effect test results show that,aqueous Rhizoma Arisaematis new processed products has obvious analgesic effect,thw effective is smaller than raw materials,but stronger than the Pharmacopoeia method processed products;The sedative anticonvulsant experiment results show that,the Arisaema new processed products has obvious sedative and anticonvulsant effects,and it is better than the Pharmacopoeia method processed products;(4)The moisture,ash and water soluble extract content of Arisaema new processed products is assaied,and the order is 9.63%,2.95%,15.54%and the content of total flavonoids content was 0.230%.Conclusion:After the improvement and optimization,the new processing technology of Rhizoma Arisaematis is able to reduce or eliminate the Arisaema toxicity and irritation,it is achieved the target of toxicity reducing and efficacy enhancing.And the operation of the processing process is simple,scientific and rational,stable and feasible,it could be used to guide the Araceae medicinal processing production. |
