| The Homalium paniculiflorum is tropical and subtropical endemic medicinal belongs to Homalium genus of Flacourtiaceae.Homalium paniculiflorum is a traditional Chinese folk medicine,used for bleeding and treatment of rheumatism?gonorrhea?sore-toxin.Under the bioassay guided,The active ingredients of the stem of H.paniculiflorum were studied through the methods of extraction,separation,purification and structural characterization.At the same time,in order to find the strong anticancer activity compounds,the anticancer activity of the crude extracts and the monomeric compounds have been evaluated,which will provide a theoretical basis and foundation material for the development and utilization of the H.paniculiflorum.Under the bioassay guided,the petroleum ether and ethyl acetate fractions of the sterm of H.paniculiflorum were isolated by column chromatography on Sephadex LH-20,silica gel and HPLC,and identified by spectroscopy of 1D-NMR,2D-NMR,MS,UV,IR,CD,X-ray and chemical methods.Thirty-nine compounds were isolated and puried from the stem of the H.paniculiflorum,including 8 new compounds.More than 7 structural types were found,including 8 aromatic glucosides,8 lignins,4simple phenylpropanoids,4 isocoumarins,4 xanthones,6 simple benzene derivatives and 5 other compounds.Compounds 1,2,6-20,23-31,35 and 36 were isolated from the Homalium genus for the first time.All compounds were isolated from Homalium paniculiflorum for the first time.Evaluated cytotoxic activity for all extracts and the monomeric compounds by MTT.The results showed that all extracts exhibited significant effects on human cancer cell lines.Compounds 30 and 31 showed that significance cytotoxicities against various human cancer cell lines?HL-60,SMMC-7721,A-549,MCF-7,SW480?in the range of IC500 at 4.08–24.14?M.Using trace dilution of monomer metabolites antibacterial tests,Compounds 25and 26 showed that significance Escherichia coli and Staphylococcus aureus,MIC50values is 1.25?M. |
