Objective This study aimed to investigate the protective effects of total flavonoids from Smilax glabra(TFSG)in weaning rats with lead poisoning by observing its influence on renal function and renal oxidative damage parameters;to explore the protective mechanism of TFSG on the kidney of weaning rats with lead induced damage by observing its influence on mRNA and protein expressions of NF-?B/I?B/IKK signal pathway and Caspase-3.Methods From adult stage Sprague-Dawley rats,male and female each half.1:1 mated successfully,female rats were randomly divided into normal control group,model group,the normal group,4 of which,model group 20.After childbirth female rats,pick male pups after weaning,maternal death.The model group were randomly divided into model group,positive control group DMSA(DMSA 70 mg/kg),low-dose,medium-dose and high-dose TFSG groups(50,100,200 mg/kg)of 5 per group 10.In addition to the normal control group was fed with deionized water,the other group was to continue to give 0.5%lead acetate solution until the end of the experiment.The model group of animals orally given distilled water,and the remaining animals were orally given different test substances.The whole experiment lasted 28 days.After the end of this experiment,the contents of serum uric acid(UA),creatinine(Cr)and blood urea nitrogen(BUN)content,the activities of renal superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px)activity and glutathione(GSH),as well as the levels of renal malondialdehyde(MDA)and reactive oxygen species(ROS)were detected according to the gulidelines of kits.In addition,determination of the lead in blood,urine and different tissues,and histopathological observation were done through renal biopsy on each group.Simultaneously,the mRNA and protein expressions of renal NF-?B/I?B/IKK pathway and Caspase-3 were detected by RT-PCR and Western blot methods respectively.Results By contrasting the normal control group and model group,the body weight ratio decreased significantly(P<0.05);UA,Cr,BUN,urine,blood,hippocampus,liver,renal lead concentrations were significantly increased(P<0.05);SOD,CAT,GSH-Px activity and GSH levels in renal tissue of rats were significantly lower(P<0.05),MDA,ROS levels in renal tissue of rats were significantly increased(P<0.05);renal of rats in model group tubular structural damage,apoptosis trends;increased significantly in the model group rat kidney NF-?B/I?B/IKK pathway and Caspase-3 mRNA and protein expression(P<0.05).Compared with model group,TFSG could significantly influence the body weight ratio in rats(P<0.05);UA,Cr,BUN,blood,hippocampus,liver,ranal lead concentration decreased significantly(P<0.05),urine lead were significantly increased(P<0.05);SOD,CAT,GSH-Px activity and GSH levels in renal tissue of rats were significantly increased(P<0.05),MDA,ROS levels in renal tissue of rats were significantly lower(P<0.05);which,in high doses TFSG can significantly improve the pathological changes in renal tissue of rats with lead poisoning;renal NF-?B/I?B/IKK pathway and Caspase-3 mRNA and protein expression was significantly decreased(P<0.05).Conclusion TFSG could significantly prevent the renal oxidative damage in weaning rats with lead poisoning,and could regulate NF-?B/I?B/IKK signal pathway by ROS.It has broad application foreground in treating the lead poisoning.
