Study On The Protective Mechanism Of Qizhi Capsule On Ischemic Stroke By Bax And Bcl-2,Regulatory Factors Of Apoptosis

Objective:This research adopts the rat model of cerebral ischemia-reperfusion,by detecting the changes of apoptosis regulating factor Bax and Bcl-2 in rat hippocampal neurons,Deeply and systematically study the clinical wide application,and have a research base of Qizhi capsule protective effect on ischemia reperfusion injury model in rat brain,provide the experimental basis for the development of new drugs for ischemic stroke,and broaden the research ideas of prevention and treatment of ischemic stroke.Method:Preparing rat middle cerebral artery occlusion(MCAO)model by using Zea Longa methods,and observing the morphological changes of brain through optical microscope,detecting the changes of Bax and Bcl-2(apoptosis regulatory factors)in hippocampus neuron on the model rat of cerebral ischemia reperfusion at different time points by immunohistochemistry detection.Results:The use of TCM Qizhi capsule early intervention can reduce neurological diseases and brain tissue morphology lesions at rats of ischemic stroke.The expression level of Bax in model group were higher than that of the dual-purpose drug group,Bcl-2 expression level were lower than dual-purpose drug group.Compared with model group,the expression of the two gene in traditional Chinese medicine group was significantly different(P<0.05).In sham operation group,western medicine group and traditional Chinese medicine group,the ratio of Bcl-2/Bax were higher than that of the model group,and the traditional Chinese medicine group compared with the model group had obvious difference.Conclusion:TCM Qizhi capsule early intervention can downregulate the expression of rat Bax of ischemic stroke,and up regulation of Bcl-2 expression,and increase the ratio of Bcl-2/Bax,inhibit the cell apoptosis of ischemia reperfusion injury.Thus the cerebral protective effect on ischemic stroke.
