| Objective: to explore the anti-inflammatory mechanism of zhuang medicine medicated thread moxibustion combined with collateral-puncturebleeding for the treatment of gouty arthritis.Methods: 13 male rats were randomly selected from 78 male rats as blank group.After being made acute gouty arthritis rat models According to Coderre Ways classic model,65 male rats were randomly divided into 5 groups: model group,western medicine group,observation group 1,observation group 2 and observation group 3,13 in each group.Rats in the blank group were not subjected to modeling and untreated.model group were untreated after modeling.The other 4 groups were given treatment after modeling: western medicine group were treat with colchicin by lavage,twice a day;the observation group 1、the observation group 2 and the observation group 3 were treated with pricking blood therapy of Zhuang medicine combined with medicated thread moxibustion respectively at different blood volume and dose of point moxibustion,once a day;the observation group 1 bloodletting 0.1ml ± 0.04 ml,point moxibustion once,Observe group 2 bloodletting 0.2ml ± 0.04 ml,point moxibustion twice,Observe group 3 bloodletting 0.3ml ± 0.04 ml,point moxibustion three times.Observe the changes of rats’ gait as well as the circumference of its joint swelling and histopathological changes.Using enzyme-linked immunosorbent assay(ELISA)detection methods,inflammatort factors such as IL-1、IL-2、IL-4、IL-6、IL-10、TNF-α、IFN-γ expression changes was observed in rat serum and articular of every group.Using statistically software to analyze the data.Result:(1)colchicine and three kinds of therapeutic dose of Zhuang medicine medicated thread moxibustion combined with collateral-puncturebleeding significantly improve the gait of rats(P<0.05),curative effect of western medicine group and observation group3 is the best.(2)colchicine and pricking blood therapy of Zhuang medicine combined with medicated thread moxibustion respectively at different blood volume and dose of point moxibustion can inhibit acute gouty arthritis joint swelling in rats,joint circumference in each treatment group compared with the model group that has differences(P<0.05),the curative effect of group 3 is the best among the 4 treatment groups(P<0.05).(3)Observation group 3 and western medicine group have equivalent efficacy in restraining the secretion of inflammatory factors such as IL-1、IL-6、IL-2、IFN-gamma and TNF alpha,promote the release of anti-inflammatory factors such as IL-4 IL-10 in the articular synovial and serum of rat model with acute gouty arthritis.Conclusion:(1)The treatment of zhuang medicine medicated thread moxibustion combined with collateral-puncture-bleeding has definite therapeutic effect on acute gouty arthritis rat models;(2)each treatment group has equivalent efficacy on improving acute gouty arthritis rats model gait,the third kind of therapeutic dose of Zhuang medicine medicated thread moxibustion combined with collateral-puncture-bleeding achieved consistently curative efficacy as colchicines;(3)zhuang medicine medicated thread moxibustion combined with collateral-puncture-bleeding of different treatment quantity to varying degrees to inhibit acute gouty arthritis joint swelling at 12h、24h、48h、72h model establishment,the efficacy of bloodletting 0.3ml±0.04 ml and point moxibustion three times is the best;(4)The treatment of zhuang medicine medicated thread moxibustion combined with collateral-puncture-bleeding effectively restrain the secretion of inflammatory factors such as IL-1 IL-6,IL-2 IFN-gamma and TNF alpha,promote the release of antiinflammatory factors such as IL-4 IL-10 in the articular synovial and serum of rat model with acute gouty arthritis,its curative effect is related to therapeutic dose;(5)This experimental study found that zhuang medicine medicated thread moxibustion combined with collateral-puncture-bleeding therapy has the function to reduce pain and tumescence and anti-inflammatory,providing theoretical guidance and reference basis for clinical application. |
