| Objective:To investigate inhibitory effects of Ampeloptin(Amp)on the proliferation of human bladder cancer cell line SW780 in vitro and to explore the apoptosis induction mechanism involved.Methods:Inhibitory effects of Amp on SW780 proliferation were determined using MTT method at 48th hour and 72th hour respectively.Apoptosis status was observed under Fluorescence Microscope.Mitochondrial membrane potential,pH value,and calcium concentration of SW780 treated with Amp at graded concentration were detected under Laser Scanning Confocal Microscope.Expression levels of p-Bcl-2,Bax,and Cleaved caspase-3 of SW780 treated with Amp at graded concentration,and at 150 μg/ml alone at 12th hour,24th hour,and 48th hour respectively,were determined using Western Blotting.Results:Amp inhibited the proliferation of SW780 significantly at the concentration of 60,72,87,104,125,and 150 μg/ml,with IC50 value of 83.91 ±2.15 and 66.52±3.60 μg/ml at 48th hour and 72th hour respectively.Apoptosis rate of SW780 treated with Amp was increased in positive correlation with the increase of Amp concentration.Compared with control group,mitochondrial membrane potential of SW780 decreased,and calcium concentration increased with the increase of Amp concentration,dose dependently,but mitochondrial pH value was not affected by Amp.Expression levels of p-Bcl-2 was decreased,Bax and cleaved caspase-3 increased in a dose-dependent and time-dependent way.Conclusion:Ampeloptin(Amp)inhibites the proliferation of bladder cancer cell line SW780 in both dose and time dependence and induces apoptosis via mitochondrial signaling pathway. |
