Salviae miltiorrhizae Radix et rhizome is the dry root or roots of Salvia miltiorrhiza Bge.It has effect of removing blood stasis,relieving pain,promoting blood circulation,tranquilizing mind and relieving vexation.Salviae miltiorrhizae Radix et rhizome has a large market of great expansion,the demands of it has increased dramatically.Wild Salviae miltiorrhizae Radix et rhizome resources is far to satisfy the need of market.With the expanding constantly of cultivation,the quality of it gradually degrades.The original traditional quality standards according to the appearance characters including size of the medicinal materials,the thickness,the length and the color no longer fit the present requirements.This research based on the modern scientific methods to establish the characteristic spectrum patterns of S.miltiorrhiza pieces from different grades,provide an ideal reference for hierarchical management of S.miltiorrhiza pieces.Through the review of the literature,Salviae miltiorrhizae Radix et rhizome authentic regions are Sichuan,Shanxi,Anhui,Shandong,Shanxi,and the best one is Sichuan,otherwise,Shanxi,Henan and Shandong are the three major areas.The medicinal materials samples from Sichuan Zhongjiang,Shanxi Shangluo and Henan FangCheng were collected and adopted.According to the requirements of the "China Pharmacopoeia" 2010 version,the different grades were processed to different batches in the factory of LiuTianBao medicinal and pharmaceutical Co.LTD(GMP authentication).The other processed samples were added,and the total was 12 onesThe 12 groups of different grades of S.miltiorrhiza pieces were as the subjects,each sample respectively was extracted with distilled water,dehydrated alcohol,chloroform,petroleum ether(60?90 ?),then,tested by the ultraviolet spectrophotometry(Appendix VA of CP 2010)in 400?200 nm wavelength range with UV scanning.The results revealed that the peaks place and patterns were similar in some extent,but the absorbance of some compounds was different relatively.According to the results,we can infer that ultraviolet absorptive components in the different S.miltiorrhiza pieces are roughly the same,however,the contents are not parallel.Thus,the difference of the ultraviolet spectrum patterns which is made by ultraviolet scanning method provides a certain reference for further classification research.HPLC was used to research the water-soluble constituents,lipid-soluble constituents and the total constituents of 12 different S.miltiorrhiza pieces,and the HPLC fingerprint patterns was established.The repeatability,stability,and characteristic were optimal.According to the water-soluble constituents,lipid-soluble constituents and the total constituents of 12 groups S.miltiorrhiza piece samples had no quantitative difference,but the peak intensity was dissimilar.As level was lower,Salvianolic acid B peak area was decreased gradually,Tanshinone ?A peak area was increased gradually,the peak areas of other components had no relationship with difference of grades.Consequently,the HPLC fingerprint patterns feature provide an reliable basis for classification research of S.miltiorrhiza pieces.
