Research The Influence In Expression Level Of HIF-1α And LDH MRNA Intervened By Chaihuang Weikuining Decoction In A Model Of Gastric Ulcer Rat With Liver Constraint And Spleen Deficiency Syndrome

Objective:To observe the expression level of gastric tissue hypoxia inducible factor alpha(HIF-1α)and lactate dehydrogenase(LDH)genes in gastric ulcer rats with liver constraint and spleen deficiency syndrome;To investigate effect and mechanism of Chaihuang Weikuining decoction’ intervention influence on these genes expression.Methods:Construct the model of gastric ulcer rats with liver constraint and spleen deficiency syndrome by multiple factors compound impacted method.In order to validate the model,rats were observed about their general physiological conditions,gastric antrum HE staining were used to observe pathological changes of the gastric tissue,using enzyme linked immunosorbent assay(ELISA)for detection content of plasma 5-serotonin(5-HT)and norepinephrine(NE)levels,and content of serum gastrin were detected by immunoradiometric assay(GAS),serum amylase activity(AMY)were detected by Chemical Colorimetry.After intervened treated by Chaihuang Weikuining decoction 14 day,expression levels of HIF-la and LDH mRNA in models’ stomach tissue were determined by Real-Time PCR technique.Results:The serum AMY,serum GAS and plasma 5-HT in model group of gastric ulcer rats were significantly decreased than those in the normal group(P<0.01),and plasma NE levels were significantly higher than those of the normal group(P<0.01),so it can be considered that the model was satisfied with the detection indicator of gastric ulcer rat with liver constraint and spleen deficiency syndrome,and it appeared significantly high expression levels of LDH of HIF-1α gene in model rats(P<0.01).After 14 days’ treatment with Chaihuang Weikuining decoction,the expression levels of LDH of HIF-1α gene appeared decreasing in varying degrees.While the decreased trend of LDH gene was more obvious,there was significant difference compared with the model group(P<0.05);Then the HIF-1α gene expression level was also appeared decreased,but there was no statistical differentiation compared with model group(P>0.05).Conclusion:The prescription of Chaihuang Weikuining decoction have different degrees’ intervention effect on expression levels of HIF-1α,LDH gene in model gastric ulcer rat with liver constraint and spleen deficiency syndrome.It may be one of action ways of Chaihuang Weikuining decoction.
