Objective To observe the expression of serum malondialdehyde(MDA),total-superoxide dismutase(T-SOD)and catalase(CAT)in gastric ulcer rats with liver constraint and spleen deficiency syndrome;To explore the mechanism of oxygen free radical and the therapeutic effect of Chaihuang Weikuining Decoction.Methods The 60 male SD rats were randomly divided into normal group,gastric ulcer group,model group,Huangqi Jianzhong decoction group,Chaihuang Weikuining decoction high-dose group,Chaihuang Weikuining decoction medium-dose group,10 rats in each group.Constructing the model of gastric ulcer rats with liver constraint and spleen deficiency syndrome by multi-factors compound simulation of TCM etiology and combined with acetic acid burning methods after adaptability of feeding 7 days.After the modeling,normal group rats,gastric ulcer group rats and the model group rats daily gavaged each 3ml normal saline,traditional Chinese medicine groups’ rats in per group each day gavaged each by decoction including crude respectively were 2.16g,3.36g,4.48g.Six groups were continuously gavaged for 14 days.Then,all rats were performed laparotomy after fasting for 24 hours,the gastric antrum was taken for pathological observation,blood was taken to detect the relevant indicators.Observing the general physical state of rats in the process of experiments;Preparing the histopathological sections of gastric antrum and dealing with hematoxylin-eosin staining,and then,observing the changes with optics microscope;To verify the reliability of the models,we used the Enzyme-labelled immunosorbent assay(ELISA)to detect the assay of amylase(AMY),gastrin(GAS)and plasma serotonin,norepineohrine(NE);To detect the assay of MDA by thiobarbituric acid method(TBA),detecting the T-SOD expression by hydroxylamine method and calculating the CAT vigour by method of visible light.Results 1.Compared with the normal group,the serum AMY,serum GAS and plasma 5-HT in model group of gastric ulcer rats with liver constraint and spleen deficiency syndrome which were constructed by multi-factors compound simulation of TCM etiology combined with acetic acid burning method were conspicuous decreased(P<0.01),and the plasma NE level was arresting increased(P<0.01),and this results above could be used to verify successful modeling;2.The assay of MDA in model group appeared high expression,whereas lower expression in T-SOD and CAT;The content of serum MDA became lower than the previous,T-SOD and CAT acticity turned into increasing after a 14-day therapy course of Chaihuang Weikuining Decoction.Conclusion 1.The model rats of gastric ulcer with liver constraint and spleen deficiency syndrome can be builded successfully by multi-factors compound simulation of TCM etiology combined with acetic acid burning methods.2.This model showed high expression of oxygen free radicals;3.The varying degrees of accommodate in the expression of MDA,T-SOD and CAT may be one of the effective ways of Chaihuang Weikuining in treating this pattern of syndrome disease.
