Extraction And Purification Of Active Components In Leaves Of Eucommia Ulmoides

Eucommia ulmoides is a kind of Eucommia deciduous tree,which is the traditional herbal medicine in china decocting the eucommia bark as medicine and having the function of strong bones,nourishing liver and kidney,reduce blood sugar,enhance immunity,anti-aging.In this paper,four kinds of active components in Eucommia ulmoides leaves(chlorogenic acid,geniposidic acid,aucubin,flavonoids)were extracted and purified.The extraction by ethanol was researched by single factor(ethanol concentration、temperature、ultrasonic power and time)test and response surface analysis,and the purificationwas carried out by chitosan precipitation,activated carbon adsorption and membrane separation.Some significant results were obtained as follow:1.When extract the active components from Eucommia ulmoides leaves with ethanol,it was found that the concentration of ethanol,ultrasonic power,temperature and time had no significant effect on the extraction ratio of geniposide and chlorogenic acid,but affect the extraction ratio of flavonoids and aucubin.Single factor and response surface analysis shown the optimum extraction conditions of four active components were:the solid-liquid ratio was 1:10(g/ml),the ethanol concentration is 48%,the extraction temperature is 48 oC,the ultrasonic is 150 W and the time is 20 min.According to the optimum extraction conditions,the extraction rate of total flavonoids was 7.25%,the aucubin was 5.65% and the chlorogenic acid was 1.51%.which were similar to the actual value.2.When extract the active components from Eucommia ulmoides leaves with enzymation,ultrasonic can promote the extraction ratio of geniposide(add by 35%),it had no significant effect on the extraction ratio of chlorogenic acid and flavonoids,but inhibite the extraction ratio of aucubin.Take into account,The reasonable conditions for enzymatic extraction were: the ratio of solid to liquid is 1:10,the temperature is50oC,the ultrasonic is150 W,the time is 10 minand the amount of pectinase is 0.5mg/g.In this condition,theextraction rate of total flavonoids was 5.04%,the aucubin was 3.21%,the chlorogenic acid was 1.76% and the geniposidic was 0.72%.3.The effective purification process as follow: firstly mixed extraction solution with 1% chitosan at 50 oC for 30 min and stood for 60 min,then decolorizated with 1% activated carbon at 50 oC for 50 min,and finally purified by 10 KDal ultrafiltration membrane.The decolorization rate could up to 95%.
