Hepatitis E(HE)is a viral hepatitis caused by hepatitis E virus(HEV).HEV is a single stranded positive strand RNA virus with a full length of 7.2 kb and contains three open reading frames(ORF).The ORF2 is responsible for encoding the unique capsid protein of hepatitis E virus,which contains 660 amino acids.Proteins are involved in viral assembly,immunization and viral cell interactions.In recent years,HEV has gradually become one of the leading causes of acute viral hepatitis worldwide.Pregnant women and the elderly are the major high risk groups.Hepatitis E cases of mortality is about 0.2 to 4%,pregnant women infected with HEV mortality can be as high as 10 to 25%.In this study,CD20/CD27/IgG/p239 as a specific marker,screening a donor in vivo HEV antibody titer inoculated hepatitis E vaccine volunteers,the separation of peripheral blood,to identify hepatitis E virus virus particles P239 specific B cells.10 monoclonal antibodies against HEV were obtained by single cell RT-PCR.Among them,10 antibodies can be better to identify HEV antigen.And the T183 human antibodies,with a high neutralization activity.In this study,HEV human antibody screening platform was successfuLly established.The platform was applied to flow cytometry and single cell PCR.It was possible to obtain pathogen-specific human monoclonal antibody in the shortest 28 days.For the prevention and control of new infectious diseases Outbreak is of great significance.With the deepening of the study of tumor immunotherapy,the application of human monoclonal antibody has been continuously expanded and the market demand has been gradually increased.The establishment of this platform has laid a good foundation for the next stage of tumor therapy research.
