Objective: By improving the excessive oxidative stress caused by obesity through exercise and diet intervention,this experiment aims to explore the effect of oxidative stress on reproductive function and its mechanism,to provide experimental basis for revealing the role of energy balance in the regulation of reproductive function,and to provide theoretical basis for exploring the means of preventing obesity through exercise/diet and the means of promoting male reproductive health.Methods: 17 mice were randomly selected as the normal control group from 54 weaned male C57BL/6J mice according to body weight,and fed with normal diet.The remaining 37 mice were the obesity group and given high fat diet for 12 weeks to establish the mouse model of obesity.After 12 weeks,the 17 normal mice were divided into two groups according to their body weight: the normal diet control group(NC,N=8),and the normal diet exercise group(NE,N=9).Of the 37 mice for the obesity model,34 survived.They were divided into four groups: the high fat control group(HC,N=8),the high fat exercise group(HE,N=9),the high fat to the normal diet control group(HN,N=8),and the high fat to normal diet exercise group(HNE,N=9).The groups of NE,HE and HNE were given 8-week treadmill training(6 days/week,1 hour/day).In the groups of HE and HNE,there was one mouse that fails to survive respectively.And then the mice were measured with their serum testosterone by ELISA method,MDA and T-SOD of testis by spectrophotometric method,and m RNA transcription and protein expression of P38 MAPK,NF-ΚB in testis by real-time PCR and Western blot methods.Results: 1.(1)Compared with the NC group,the body weight,the abdominal fat weight and the Lee’s index of HC group were significantly increased(P<0.01);(2)Compared with the HC group,the above indexes of the HE,HN and HNE groups were significantly decreased(P<0.01).2.(1)Compared with the NC group,the testis coefficient and serum testosterone of HC group were significantly decreased(P<0.01);(2)Compared with the HC group,the serum testosterone of the HE group was significantly increased(P<0.01),the testis coefficient of the HN group was significantly increased(P<0.05),and the above two indexes of the HNE group were significantly increased(P<0.01).3.(1)Compared with the NC group,the testis MDA of the HC group was significantly increased(P<0.01)and the T-SOD and SOD/MDA of the HC group were significantly decreased(P<0.01);(2)Compared with the HC group,the testis MDA of the HE,HN and HNE groups was significantly decreased(P<0.01),the T-SOD and SOD/MDA of the three groups were significantly increased(P<0.01).4.(1)Compared with the NC group,the m RNA and protein expression levels of P38MAPK、the P-p38 MAPK and the NF-ΚB in the testis of the HC group were significantly increased(P<0.01);(2)Compared with the HC group,the above indexes of the HE,HN and HNE groups were significantly decreased(P<0.01).Conclusion: 1.The 8-week high-fat diet leads to fat accumulation,enhances the level of oxidative stress,upregulates the ROS-P38MAPK-NF-ΚB signaling pathway in testicular tissue,and decreases the serum testosterone levels.It is one of the causes of reproductive potential decrease.2.The 8-week treadmill exercise,the diet intervention and the combination of the two decreases the oxidative stress level,regulate the inhibited ROS-P38MAPK-NF-ΚB signaling pathway down in testicular tissue,and increase the serum testosterone and the male reproductive potential.
