Objective:To study the effect of ethyl acetate fractions of Ferula sinkiangensis node invasion and migration ability of HCT116 cells.To investigate the anti-tumor effect of mice with S-180 solid tumor,CT-26.WT ectopic allograft and orthotopic transplanted mice.Methods:1.CCK-8(Cell Counting Kit-8 kit)was used to detect the inhibitory effect of Ferula sinkiangensis on the proliferation of HCT 116 cells,and the half inhibitory concentration of Ferula sinkiangensis was calculated.(IC50),cell scratch test and Transwell assay was used to observe the migration and invasion of HCT116cells.2.The establishment of mouse S-180 solid tumor model,60 mice were randomly divided into normal control group,tumor model group,low,middle and high dose group of Ferula sinkiangensis group.The mice were administered with the ethyl acetate part 10 days.The organ index,body weight and tumor inhibition rate were observed in mice.The levels of serum IL-2,IFN-γand TNF-αwere measured by ELISA 3.Using CCK-8 assay in vitro Ferula sinkiangensis ethyl acetate extract on CT-26.WT cell antitumor activity,cellular proliferation inhibition rate and half inhibitory concentration(IC50);the detection of Ferula sinkiangensis total apoptosis rate of CT-26.WT cells by flow cytometry;4.Establishment of CT-26.WT ectopic tumor mouse model,random the BALB/c mice were divided into normal control group,tumor model group,ferula sinkiangensis ethyl acetate fractions is low,medium and high dose group,cisplatin group,organ index,body weight of mice,observe the pathological section of ferula sinkiangensis group gavage after administration of 10days,the tumor inhibition rate was calculated.5.Establishment of CT26.WT in situ transplanted mouse model,60 BALB/c mice were randomly divided into normal control group,model group,Ferula sinkiangensis tumor of low medium and high dose group,cisplatin group,organ index,body weight of mice and the pathological changes of tumor were observed after administration of 10 days,the tumor inhibition rate was calculated and ELISA method.The levels of IL-12p70,IFN-γ,TNF-α,IL-6,IL-10 and MCP1 in serum were measured by cytometric bead array(CBA).Results:1.With the increase of medicine concentration,OD value in Ferula sinkiangensis resin group and cisplatin group decreased gradually;inhibition ratio and medicine concentration were dependent;IC50 was 43.98μg/mL in Ferula sinkiangensis resin group and 36.77μg/mL in cisplatin group.Compared with control group,affected by separation parts of ethyl acetate from Ferula sinkiangensis resin,migration and invasion abilities of HCT116 cells were weakened significantly(P<0.05,P<0.01).2.Low,medium and high dose group of Ferula sinkiangensis,the tumor inhibition rate was 40.43%,50.86%,58.20%.Ferula sinkiangensis dose group of S-180 mice spleen index were significantly increased,there was significant difference compared with the control group(P<0.05),thymus index and weight compared with the normal control group had no obvious change.Ferula sinkiangensis in high dose group was significantly increased in S-180 mice IL-2,IFN-γ,TNF-αlevel,compared with the model group,the difference was statistically significant(P<0.05),each dose group significantly increased levels of TNF-α,compared with the model group,the difference was statistically significant(P<0.05).3.The IC50 of the ethyl acetate part of Ferula sinkiangensis was 28.23,and cisplatin was 4.81,Flow cytometry test Ferula sinkiangensis 125μg/mL concentration of CT-26.WT cell apoptosis rate was80.3±12.6.4.Ferula sinkiangensis low,medium and high concentrations of ferula on CT-26.WT solid tumor mice tumor inhibition rate(38.26±7.82)%and(48.60±9.09)%and(66.54±12.02)%,and the rate of inhibition of cisplatin(95.18±3.02)%,middle and high dose group of mice spleen index were increased significantly,compared with the normal control group(P<0.05),compared with the model group,the difference was statistically significant,thymus index and weight compared with the normal control group had no obvious change.5.Ferula sinkiangensis ethyl acetate fractions of ferula CT-26.WT mice orthotopic transplantation tumor inhibition rate(38.26±7.82)%and(48.60±9.09)%and(66.54±12.02)%,and the rate of inhibition of cisplatin(95.18±3.02)%,middle and high dose group of mice spleen index obviously,compared with the normal control group(P<0.05),compared with the model group,there were statistically significant differences in body weight,compared with the normal control group had no obvious change.6.The level of IL-12p70,TNF-α,IFN-γ,IL-10 and MCP1 in serum of CT-26.WT in situ transplanted tumor mice increased and IL-10 level decreased.Conclusion:1.The isolation of ethyl acetate in Ferula sinkiangensis has a certain inhibitory effect on the proliferation,migration and invasion of human colon cancer cell line HCT116 Tumor.2.The level of IL-2,IFN-γand TNF-αin serum of S-180 solid tumor mice was inhibited by Ferula sinkiangensis,which could inhibit the tumor of S-180 solid tumor mice.3.Ferula sinkiangensis can inhibit the proliferation of CT26.WT cells in vitro,and inhibit the growth of CT-26.WT ectopic xenografts in vivo.4.Ferula sinkiangensis on CT-26.WT in situ transplanted mice have a certain role in inhibiting tumor growth.
