Part 1:The expression of MGMT gene in non-small cell lung cancer researchObjective:Testing the expression of MGMT gene in Non-small cell lung cancer tissue and tissue adjacent to carcinoma and finding the relation with clinical characteristics of non-small cell lung cancer.Method:Collect 43 patients with non-small cell lung cancer surgery for lung cancer tissue and tissue adjacent to carcinoma from the first affiliated hospital of guangxi medical university.The RNA of non-small cell lung cancer tissue and tissue adjacent to carcinoma were extracted with trizol method,which takerealtime fluorescent quantitative PCR to detection the gene expression of MGMT in lung cancer tissue and tissue adjacent to carcinoma.Non-small cell lung cancer tissue expression and the expression of tissue adjacent to carcinoma compared with paired t-test,the relative expression quantity relation with clinical indicators with chi-square test and survival analysis using Kaplan Meier-survival analysis.MGMT gene in non-small cell lung cancer survival rates have no obvious difference between different expression level in18 months(?~2=0.188,P=0.664).Result:MGMT gene expression in non-small cell lung cancer tissue was lower than that tissue adjacent to carcinoma(t=3.065,P=0.005,P<0.05),at the same time,the expression of MGMT gene and gender(?~2=13.810,P=0.003),age(?~2=20.419,P=0.000),pathology types(?~2=15.326,P=0.002)and clinical stage(r=-0.429,P=0.026,P<0.05)have obvious relationship.Conclusion:In non-small cell lung cancer tissues MGMT gene expression is obvious lower than tissue adjacent to carcinoma,and with the gender,age,pathology classification and clinical staging a clear relationship,so it can conclude that the expression of MGMT gene is small cell lung cancer the important symbol of reflection,progress and prognosis of non-small cell lung cancer diagnosis to provide a positive guiding significance,and provide a reference for future research.Part 2:MLH1 abnormal gene methylation in non-small cell lung cancer and the correlation between MGMT gene and clinical studies of the relationshipObjective:Non-small cell lung cancer tissue and tissue adjacent to carcinoma MLH1 gene abnormal methylation in contact with clinical characteristics of non-small cell lung cancer.Method:Collect 43 patients with non-small cell lung cancer surgery for lung cancer tissue and tissue adjacent to carcinoma from the first affiliated hospital of guangxi medical university.Extracted carcinoma genome DNA of non-small cell lung cancer tissues and its tissue adjacent,Using methylation-Specific PCR detect MLH1 gene methylation status in non-small cell lung cancer tissue and adjacent.Comparison between groups by chi-square test,correlation with Spearman correlation,survival analysis using Kaplan Meier-survival analysis.Result:In non-small cell lung cancer tumor tissue MLH1 abnormal methylation level higher than tissue adjacent to carcinoma(?~2=8.667,P=0.034,P<0.05),the abnormal methylation MLH1 gene and gender(?~2=12.349,P=0.006)and clinical stage(?~2=41.349,P=0.000)has obvious relationship.After the comparison with the MGMT gene found that MGMT gene expressionandMLH1abnormalmethylintonegativecorrelation(r=-0.429,P=0.026,P<0.05).MLH1 abnormal methylation in non-small cell lung cancer with survival rate has no obvious difference between methylation 18months(?~2=0.828,P=0.363).Conclusion:MLH1 gene in non-small cell lung cancer tissue abnormal methylation level obviously higher than that of tissue adjacent to carcinoma,and it has an obvious relationship with gender and clinical staging,so you can think that the abnormal methylation MLH1 gene and the occurrence,progress and prognosis of non-small cell lung cancer has important relation,the diagnosis of non-small cell lung cancer to provide a positive guiding significance,and provide a reference for future research.In addition,the lower expression of MGMT gene with the exception of MLH1 gene methylation exist obvious correlation,it shows that the two genes in cancer genetics and epigenetics of the role is not isolated,but there is a relatively complex pathways.
