Effects Of Chaiyu Dixian Decoction On Ethology,HPA Axis And Expression Of BDNF And TrkB Protein In The Hippocampus Of Perimenopausal Depressive Disorder Rats

Objective:Chaiyu Dixian Decoction was used as an intervention method,with smoothing the liver and reinforcing essence of the kidney,for teatment the Perimenopausal Depressive Disorder rat model.Then observe the changes of body weight,behavior in rats,the space exploration time of Morris water maze,the contents of serum Corticotropin Releasing Hormone(CRH),Adrenocorticotropic Hormone(ACTH),Corticosterone(Cort),and the protein expression of Brain Derived Neurotrophic Factor(BDNF)and Tyrosine Receptor Kinase B(TrkB)in hippocampus were determined.And then reveal the mechanism of Chaiyu Dixian Decoction in the treatment of Perimenopausal Depressive Disorder.Methods:1.A total of 48 female Sprague-Dawley rats(age from 3~4 months)were qualified by Open Filed Test.Randomly divided into the sham group,the model group,the medicine group,the Chaiyudixian high,medium,and low dose groups,with 8 rats in each group.The sham group underwent sham operation,while the other groups were received 28 days of Chronic unpredictable mild stress stimulation combined with lonely breeding after ovariectomy.2.To start the drug intervention after categorized,from the first day that the model establishing,the model group and the sham group were administrated with 0.9%NaCl,as well as the positive control group were administrated with Fluoxetine Hydrochloride Capsules and Estradiol Valerate and the traditional Chinese medicine groups were fed with diffenent concentrations of Chaiyu Dixian Decoction once a day respectively.During the stress process,all of rats body weight and behaviors were observed by tests,and the abilities of rats to learn and identify the space were evaluated by Morris water maze.3.After 28 days of administration,the related indexes were detected after behavioral tests were completed:①Observing the CRH,ACTH and CORT level in serum of rats by ELISA.②Measuring the expression of BDNF and TrkB protein in the hippocampus of rats by WB.Results:1.Behavior indexes:During the modeling period,compared with the sham group,the body weight of rats in the model group were increasing slowly,the scores of Open Field Test and Sucrose Preference Test were significantly reduced(P<0.01);the space exploration time of Morris Water Maze and the Forced Swimming time were significantly prolonged(P<0.05,P<0.01);Compared with the model group,the rest’s body weight,ethology,space exploration time of Morris water maze were varied(P<0.01,P<0.05).2.Results of CRH,ACTH,and CORT level:Compared with the sham group,the results of Elisa test showed that the content of CRH,ACTH,and CORT in serum were increased(P<0.01).Compared with the model group,all the rats in each administration group could significantly reduce the elevated content of CRH,ACTH and CORT in serum(P<0.01,P<0.05).3.Results of the BDNF,TrkB expression by WB:The BDNF,TrkB protein expression of the model group were lower than the sham group(P<0.01).Compared with the model group,the expression of BDNF,TrkB protein in hippocampus of rats treated with each drug were increased to varying degrees(P<0.05,P<0.01).Conclusion:1.Through Chronic mild unpredictable stress for 28 days after ovariectomy method may provide a valuable animal model of Perimenopausal Depressive Disorder(PDD).The model simulated the incidence of PDD in the ethology and behaviors.And provide a reliable experimental model for further study of the pathogenesis of PDD.2.Chaiyu Dixian Decoction,which has the effect of smoothing the liver and reinforcing essence of the kidney,can obviously regulate the depression-like behavior of the rats with depression in the perimenopausal period,regulate the hyperactive HPA axis,increase the expression of BDNF and TrkB protein in the hippocampus,and play the role of anti-menopausal depression.
