Anti-obesity,Lipid-lowering Effects And The Clinical Application Of Ethanol Extract Of Vernonia Amygdalina Leaves

Obiectives:Bitter leaves(Vernonia.amygdalina leaves)are widely used by the people in Africa,Southeast Asia,Taiwan and other areas as an antineoplastic herbal medicine.In this current study,we for the first time to investigate the effects of ethanol extracts from Vernonia amygdalina leaves on anti-obesity and lipid-lowering.Methods:C57BL/6 mice,were randomly divided into 4 groups,8 mice in each group:normal control group(normal),high fat diet group(HFD),orlistat group(HFD+O),ethanol extracts from Vernonia amygdalina leaves group(HFD+V).Except for normal control group,the other groups were given high fat diet.In the HFD+O group,orlistat was administered at 100mg/kg body weight at the same time of high fat diet,and the HFD+V group was treated with the ethanolic extract from Vernonia amygdalina leaves at 100mg/kg body weight at the same time of high fat diet.The above diet and medication for 20 weeks,the general status of each animal and animal food intake were recorded;weighed once a.week.Empty body weigh and length of mice were measured at the endpoint of the experiment,BMI index were calculated.Blood was collected by eyeball extirpating.Liver of the animals were removed and weighed.Fat tissue around mesenterium,both kidneys,bilateral epididysis was gathered and weighed.The liver index and fat coefficient were calculated.Determined the plasma TG,TC,LDL-C and liver lipid content.Partial liver from the animals were frozen in ice cold and sliced up respectively 7μ m and 10 μ m thickness sections for HE and oil red 0 staining to observe the morphology of liver and lipid droplet accumulation.Results:The animal experiment results showed that the ethanol extracts from Vernonia amygda.lina leaves can significantly depress the obese mice body weight,BMI and body fat weight;decrease in TC,LDL-C level and hepatic TG level;significantly improve the liver structure disorder induced by high-fat diet and fatty degeneration.Conclusion:Ethanolic extract from Vernonia amygdalina leaves can significantly depress the body weight of obese mice induced by high-fat diet,and can significantly regulate blood lipid.Meanwhile,the liver structure disorder and lipid droplet accumulation are also significantly improved.
