Vernonia amygdalina Del.,a member of the genus Vernonia family,which was originated in South Africa.It has high economic and medicinal value.In local area,it was traditionally used to treat malaria,fever and so on.Recently,it was reported that the Vernonia amygdalina was rich in?7,9(11)stigmatane-type steroidal saponins with complex structure and certain physiological activity.In order to further clarify the constituents of Vernonia amygdalina and find the natural products with novel structure,we studied the chemical constituents in the leaves of Vernonia amygdalina.(1)Cytotoxic and anti-inflammatory active test for Vernonia amygdalina Del.MTT method was adopted to test its cytotoxic and anti-inflammatory activity.It was found that both of the 95%ethanol and the dichloromethane parts showed cytotoxic and anti-inflammatory activities.Among them,the anti-inflammatory and cytotoxic of the dichloromethane fraction was significant,which was worthy of further study.(2)Study on the chemical composition of dichloromethane part of Vernonia amygdalina Del.The classical separation and purification methods were taken to separate and purify dichloromethane,the structure of these compounds were identified by the the combination of magnetic spectrum data(ESI-MS,1H-NMR,13C-NMR),1-8 were saponins,9 was a flavonoid and 10 was a coumarin.These compounds were identified as vernoramyosides D(1),vernoramyosides E(2),vernonioside B1(3),veramyoside J(4),vernonioside B2(5),vernonioside D2(6),vernoamyoside D(7),vernoamyoside C(8),Lutin-7-O-?-D-glucopyranoside(9),4,7-dimethoxyl-5-methyl-coumarin(10).1-2 were new compounds.All the isolates above were screened for cytotoxic and anti-inflammatory activities,however,the results showed that 1-10 had no cytotoxic and anti-inflammatory activities.Further screening experiments for their tumor cell multidrug resistance activity showed that compound 7 has obvious Multidrug resistance reversal activity.(3)Composition analysis of methylene chloride active parts of Vernonia amygdalina Del.HPLC and LC-MS/MS methods were applied to analyze the active fraction.The separated and identified compounds were used as standard materials to summarize the mass spectrometric cleavage law of?7,9(11)stigmastane-type steroidal saponins for the first time.By this method,17 compounds were identified,including 4 unknown compounds.In this paper,an activity-oriented separation method was used to study the chemical and biological activities of V.A.,the results rich of the varity of saponins laid the foundation for further research and development of lead compounds.
