| Objective:To investigate the expression of ADAM10 and p62 in HCC,and their relationship with pathological features and the prognosis of HCC,and to find out potential therapeutic target and new prognostic indicator for HCC.Methods:Collect 2013-07 to 2017-12,the First Affiliated Hospital of Jilin University in resected HCC paraffin,using immunohistochemical SP method(streptavidin biotin-peroxidase immunohistochemical method)to detect the expression of ADAM10 and p62 in pathological confirmed 93 cases of hepatocellular carcinoma and 87 cases of adjacent normal tissues.And their relationship with pathological features of HCC.Survival analyses are undertaken according to the follow-up data of the patients.Statistical processing: the data were analyzed by SPSS22.0 statistical software.According to the different objects of study,the counting data were analyzed by Pearson chi-square test,and the correlation analysis between the two used spearman grade correlation analysis method.The survival data were analyzed by Kaplan-Meier and the survival curve was drawn.The difference was tested by Log-rank method and the independent prognostic factors were analyzed by Cox risk model.The difference was statistically significant(P < 0.05).Results:The positive expression rate of ADAM10 in HCC tissues was 69.9%,while in adjacent normal tissues was 39.1%(P<0.01).There were significant associations between positive expression of ADAM10 and histological grade,tumor size,venous invasion,metastasis and TNM stage(P<0.05),but not with age and gender(P>0.05).Furthermore,ADAM10 protein expression was significantly associated with shortened survival time(P<0.01).The positive expression rate of p62 in HCC tissues was 54.8%,while in adjacent normal tissues was 31.0%(P<0.01).There were significant associations between positive expression of p62 and histological grade,tumor size,venous invasion,metastasis and TNM stage(P<0.05),but not with age and gender(P>0.05).Furthermore,p62 protein expression was significantly associated with shortened survival time(P<0.05).There was no significant correlation between the expression of ADAM10 and p62 protein in HCC(P>0.05).Multivariate Cox regression analysis showed that ADAM10 expression and vascular invasion could be independent prognostic factors for predicting survival rate of these patients.Conclusion:1.ADAM10 protein and p62 protein were strongly expressed in a large proportion of HCC cases.There were significant associations between positive expression and histological grade,tumor size,venous invasion,metastasis and TNM stage(P<0.05),but not with age and gender(P>0.05).The expression was significantly associated with shortened survival time(P<0.05).2.There was no significant correlation between the expression of ADAM10 and p62 protein in HCC(P>0.05).3.ADAM10 expression and vascular invasion could be used to predict patient survival after treatment. |
