Objective:(1)Establishment and identification of rat model of stress induced hypertensive rats.(2)Investigate the differential metabolic components in urine and serum of stress induce hypertension rats based on the method of metabonomics,and the pathogenesis of stress-induced hypertension is discussed by means of differential metabolic components to provide experimental basis for further progress of drug treatment of stress induce hypertension rats.Methods:(1).30 male Wistar rats were divided into normal control group and stress induce hypertension model group,Normal control group without any treatment while stress induce hypertension model group rats was given stimulation such as foot shock,braking,clip tail.using the rat tail arteriometer to measure each rat tails artery systolic blood pressure.collect each groups of rats plasma,liver,adrenal glands,kidneys,detect stress related indicators such as cortisol and aldosterone malondialdehyde,heat shock protein 60 and mineralocorticoid receptor by respectively using radioimmunity methods,TBA method and immunohistochemical method,determine the success of establishment of stress induce hypertensive model.(2)after the establishment of the stress induce hypertensive model,the urine and serum of rats were collected,and the differential metabolic components in urine and serum of rats were detected by nuclear magnetic resonance spectroscopy.Results:(1)The rat model of stress induce hypertensive rats was successfully established by means of the combined stress stimulation such as foot shock,braking,clip tail.Each groups Blood pressure was detected by carotid artery test,and showing that compared with normal group,the systolic and diastolic blood pressure increased in the model group,and the results were statistically significant(P < 0.05).(2)the content of stress related indicator including aldosterone,cortisol,malondialdehyde,salt corticosteroid receptor and heat shock protein 60 were increased in the plasma of stress induced hypertensive model rats,compared with normal control group differences were statistically significant(P < 0.05).(3)the content of isoleucine,leucine,valine,glycoprotein,beta-glucose,VLDL,acetic acid-acetate and carnitine in the serum sample of stress induce hypertension model group rats was increased,Compared with the control group,the difference was statistically significant(P < 0.05).the content of Tyrosine,phenylalanine,acetone and 1-methyl histidine,beta glucose,acetate and acetic acid,formic acid,acetyl acetate salt,malonic acid,citric acid,malic acid,creatinine,and urea in urines of stress induce hypertension model group rats was increased,Compared with the control group,the difference was statistically significant(P < 0.05);Conclusion:(1)The model of stress induce hypertensive rats was successfully established by means of the combined stress stimulation such as foot shock,braking,clip tail.(2)Stress induced hypertension is a metabolic disease,stress induced hypertensive rats have disorder of lipids,amino acids,glycemic metabolisms and tricarboxylic acid cycles.
