Objective:To study the expression of SOX2 and PD-L1 in breast invasive ductal carcinoma(breast cancer)and its relationship with clinicopathological parameters of breast cancer.Methods:The expression of SOX2 and PD-L1 in 239 breast cancer tissues were detected by immunohistochemical SP method.The relationship between SOX2 and PD-L1 expression and clinicopathological factors was analyzed by x2test.Spearman relation analysis method was used to determine relationship of SOX2 and PD-L1 expression.Kaplan-Meier’s method and COX regression analysis were used to analyze prognosis,and the log-rank test was used to determine their significance.Results:(1)The expression ofSOX2in breast cancer(40.17%)was significantly higher than that in normal breast tissue(8.54%,P<0.05).The positive expression rate of PD-L1 in breast cancer(35.15%)was significantly higher than that in normal breast tissue(16.43%,P<0.05).(2)The positive rate of SOX2 in patients with lymph node metastasis was significantly higher than that without lymph node metastasis of breast cancer(P<0.05),The positive rate of SOX2 in patients of stage III breast cancer was significantly higher than that of stage I/II(P<0.05),The positive rate of SOX2 in non-menopausal women was significantly higher than that in menopausal women(P<0.05).The positive rate of SOX2 of Lumina B,Her-2Positive and triple negative subtypes patients were significantly higher than those in Lumina A(P<0.05).The positive rate of PD-L1 expression in patients with lymph node metastasis was significantly higher than that without lymph node metastasis(P<0.05),The positive rate of PD-L1 in patients of stage III breast cancer was significantly higher than that of stage I/II((P<0.05).The positive rate of PD-L1 of Lumina B,Her-2 Positive and triple negative subtypes patients were significantly higher than those in Lumina A(P<0.05);(3)In 239 cases of breast cancer.there were 70 cases were both SOX2 positive and PD-L1 positive,there were 129 cases were both SOX2 negative and PD-L1 negative.The SOX2 expression and PD-L1 expression in breast cancer was positively correlated(r=0.648,P<0.05).(4)The 5-year recurrence rate of SOX2 positive group was higher than that of negative group(36.46%vs 9.09%;x2= 30.835,P<0.05);The 5-year recurrence rate of PD-L1 positive group was higher than that of negative group(45.24%vs 6.45%;x2=57.949,P<0.05),which is independent of PD-L1 positioning(the recurrence rates of PD-L1 membrane-positive,cytoplasm-positive and both were 51.9%,25.0%and 38.9%,respectively);the recurrence rate of both SOX2 and PD-L1 negative was the lowest:the recurrence rates of both SOX2 and PD-L1 negative group,only SOX2 positive group,only PD-L1 positive group and both SOX2 and PD-L1 positive were 3.1%,42.9%,50.0%and 35.7%respectively,P<0.05).(5)The 5-year mortality rate of SOX2 positive group was higher than that in the negative group(28.13%vs 3.50%;x2= 21.084,P<0.05);The 5-year mortality rate of PD-L1 positive group was higher than that in the negative group(33.33%vs 2.58%;X2=46.320,P<0.05),which is independent of PD-L1 positioning(the recurrence rates of PD-L1 membrane-positive,cytoplasm-positive and both were 37.0%,25.0%and 27.8%,respectively);the 5-year mortality rate of both SOX2 and PD-L1 negative was the lowest:the 5-year mortality rate of both SOX2 and PD-L1 negative group,only SOX2 positive group,only PD-L1 positive group and both SOX2 and PD-L1 positive were 0.8%,21.4%,34.6%and 27.1%respectively,P<0.05).Conclusion:(1)The expression of SOX2 and PD-L1 protein in breast cancer can be used as an index of breast cancer malignancy;the abnormal expression of SOX2 and PD-L1 protein is related to the development of breast cancer;(2)The expression of SOX2 in breast cancer is associated with lymph node metastasis,TNM stage,menopausal status and malignant molecular subtypes,the expression of PD-L1 in human breast cancer is associated with lymph nodes metastasis,advanced TNM stage and malignant molecular subtypes;(3)The positive expression of SOX2 and PD-L1 in breast cancer is associated with poor prognosis,PD-L1 is an independent prognostic parameter;(4)The expression of SOX2 and PD-L1 in breast cancer was positively correlated.
