Purpose: To observe the effect of Yiqi Jianpi Huashi Recipe on gastric emptying rate and small intestine advancing rate in mice with spleen deficiency according to the principle of replenishing qi,invigorate the spleen and dampness,and to observe the changes of spleen deficiency brain peptides Based on the analysis of brain intestine axis,the method of supplementing Qi and invigorating the spleen and dampness to regulate the secretion of brain-gut peptides and promote gastrointestinal motility.To observe the effect of Yiqi Jianpi Huashi Recipe on ERK signaling pathway in duodenal tissue of spleen deficiency mice and its effect on intestinal mucosal barrier in spleen deficiency mice,and to explore the mechanism of Yiqi Jianpi Moisture Method for Yiqijian Spleen and dampness method provides basis for treatment effect and mechanism of traditional Chinese medicine treatment with low gastrointestinal motility.Method: Experimental study: 40 mice were randomly divided into normal control group,model control group,positive control group,and Yiqi Jianpi Hushui Fang group.Except the normal control group,the other groups were treated with rhubarb water.The decoction gavage method was used to establish the mouse model of spleen deficiency.After successful modeling,mice in each group were given intragastric administration.The positive control group was administered with cisapride suspension.Yiqi Jianpi Huashi Decoction was administered with Jianyiqi Jianpi Huashi decoction,and the normal control group.The model control group was given an equal volume of distilled water.After 7 days of drug administration,the mice were sacrificed by cervical dislocation.The relative residual rate of the pigment in the stomach and the advancing rate of the small intestine were measured.The levels of SP and VIP in the serum of the mice were measured by radioimmunoassay.The duodenum of the model mice was observed by HE staining.Pathological morphology of the intestinal mucosa,Western Blot analysis of ERK1/2,P-ERK1/2 and Occludin protein expression in the duodenal tissue of mice.Result:The results of the experimental study showed that compared with the normal control group,the gastric emptying rate and intestinal propulsion rate in the model control group were significantly reduced,the content of SP in the mouse serum was significantlyreduced,and the content of VIP was significantly increased.HE stained sections were observed in the control group.The villus was broken,edema between the basal layer and the lamina propria occurred,and the muscular layer became thin.Western Blot showed that the expression of P-ERK1/2 in the duodenum of the model control mice increased,and the expression of Occludin decreased.Compared with the model control group,the serum SP content increased and the VIP content decreased after drug intervention.The gastric emptying rate and small intestine advancing rate of mice in the positive control group and the Yiqi Jianpi Hushui Recipe were significantly higher than those in the model control group.HE staining showed that the positive control group and the Yiqi Jianpi Huashi Recipe group had relatively complete duodenal villus and decreased edema.Western Blot showed that the expression of p-ERK1/2 was decreased in the duodenal tissue.Occludin’s Increased expression.Conclusion: The spleen deficiency mice had slower gastric emptying and small intestine advancing than the normal mice.The serum SP content decreased,the VIP content increased,the edema between the duodenal epithelial cells and the lamina propria was observed,and the intestinal villi was structurally disordered.There was breakage and shedding.The expression of P-ERK1 / 2 in the duodenal tissue increased,and the expression of Occludin protein decreased.Yiqi Jianpi Hushui Decoction can better promote the recovery of gastrointestinal motility in spleen deficiency mice with gastrointestinal motility,and its mechanism may be achieved by increasing the content of substance P in serum and decreasing the content of VIP.Changes in microscopic duodenal mucosa may be related to the suppression of ERK signaling pathways and the restoration of their tight junctions,thereby eliminating edema.
