| ObjectiveTo establish a model of nucleus pulposus cells degeneration,to verify the relationship between the expression of miRNA-125a-5p and the degeneration of intervertebral disc.detect the expression of signal transduction pathway mediated by miRNA-125a-5p in nucleus pulposus cells.The effect of astragaloside on the expression of miRNA-125a-5p and its target gene mediated signal transduction pathway was investigated by interfering with IL-1 to induce degeneration of nucleus pulposus cells,and the effect of astragaloside on apoptosis of nucleus pulposus cells was revealed.MethodsThe degeneration of nucleus pulposus cells induced by IL-1 was divided into five groups:(1)normal control group,(2)IL-1β model group,(3)IL-1β 20mg/L astragaloside group,(4)IL-1β 40mg/L astragaloside group,(5)IL-1β 80mg/L astragaloside group.Effects of Astragaloside A on Proliferation of Human Nucleus pulposus cells by CCK-8;expression of Astragaloside on miRNA-125a-5p and its Target Gene mediated signal Transduction Pathway by RT-PCR;expression of P21,Caspase-3,Bcl-2 and Bax protein in Nucleus pulposus cells detected by Western blot.ConclusionThe proliferation of human nucleus pulposus cells was affected by the concentration of astragaloside and the time of intervention.The proliferation of nucleus pulposus cells was the most obvious in IL-1β 80mg/L group after 72 hours of intervention.Astragaloside can regulate the pathway mediated by miRNA-125a-5p and its target gene is TP53INP1,promote the expression of miRNA-125a-5p,inhibit the overexpression of target gene TP53INP1,down-regulate the expression of BAK1,Bax and Caspase-3,inhibit the apoptosis of nucleus pulposus cells,and increase the concentration of astragaloside.The effect was gradually enhanced,showing a dose dependent tolerance. |
