| Objective:The purpose of this study is to investigate the expression of miRNA-4516 in the plasma and tissue of hepatocellular carcinoma(HCC)and to analyze its diagnostic evaluation value in HCC.Method:Forty HCC patients blood were collected from October 2015 to December 2016,in Nanhua Hospital Affiliated to the University of South China and the First Affiliated Hospital of University of South China.And the clinical datas were collected.At the same time,the blood samples were collected from 24 non-hepatocellular carcinoma according to the inclusion criteria and the exclusion criteria in Health examination center of Nanhua Hospital Affiliated to the University of South China.Real-time PCR was used to detect the expression of miRNA-4516 in the plasma of control group and hepatocellular carcinoma patients.Forty-four HCC patients tissue that were diagnosed by biopsy were collected from January 2014 to December 2016,in the Second Xiangya Hospital of Central South University.And the clinical datas were collected.At the same time,the paracancer tissue samples were collected.Real-time PCR was used to detect the expression of miRNA-4516 in the tissue of HCC patients tissue and paracancer tissue.Results:1.MiR-4516 in the plasma was significantly up-regulated in patients with HCC as compared with healthy control subjects(P<0.05).MiR-4516 in the tissue was significantly up-regulated with HCC tissue as compared with the paracancer tissue(P<0.05).Statistical analysis revealed that there were no significant differences in the relative expression of the patients in different age,stage,tumor size,AFP level,HBVDNA and so on(P>0.05).3.The expression of miRNA-4516 in HCC patients with AFP <400ug/l was significantly higher than that in the normal control group(P<0.05).4.The area under the ROC curve(AUC)according to the mi R-4516 expression in plasma was 0.74,and the sensitivity is 62.5% and the specificity is 83.3%.When it combined with AFP,the area under the ROC curve is 0.95,and the sensitivity is 89.7% and the specificity is 90.1%.The area under the ROC curve(AUC)according to the mi R-4516 expression in tissue was 0.83,and the sensitivity is 76.7% and the specificity is 78.3%.When it combined with AFP,the area under the ROC curve is 0.94,and the sensitivity is 91.4% and the specificity is 87.5%.5.Kaplan-Meier showed that the HCC patients with high expression levels of miRNA-4516 have lower survival duration than those with low expression levels(P<0.05).Conclusion:Detecting miRNA-4516 can be used as a useful diagnostic and prognostic biomarker for HCC which may help to diagnose HCC.AFP is the most commonly used laboratory test for the diagnosis of HCC.However,there are some false negatives or false positives in clinical application.Combined with miRNA detection,AFP can improve the clinical diagnosis rate,reduce missed diagnosis,and provide the possibility for early diagnosis. |
