Objective:The purpose of experiment was to detecte the expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 in colorectal carcinoma(CC),colorectal adenoma(Ad),colorectal normal mucosa(NM)as well as the correlation of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 in colorectal cancer and the relationship between clinic pathological indicators,exploreing the meaning of various factors in early diagnosis and targeted treatment of colorectal cancer.Method:This experiment used immunohistochemical(IHC)observed IGF-1,IGF-1R,IGFBP-3 and GLUT-4 protein staining in three groups of different colorectal tissue,using light microscope to judge results.Western blotting(WB)was used to t examine the expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 protein in three groups of different colorectal tissue and qReal-time PCR(RT-PCR)was used to the examine the expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4mRNA in three groups of different colorectal tissue.Results:1 Results of Immunohistochemistry1.1 The expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 protein in CC,Ad and NMThe positive rate of IGF-1,IGF-1R and GLUT-4 was increased gradually in NM,Ad,CC,the difference among groups was statistically significant(P<0.01),The positive rate of IGFBP-3 was reduced gradually in NM,Ad,CC,the difference among groups was statistically significant(P<0.01).1.2 The relationship between clinic pathological indicators and the expression of IGF-1,IGF-1R,IGFBP-3,GLUT-4 in CCThe expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 has no connection with age,sex and differentiation in CC(P>0.05).The protein expression of IGF-1,IGF-1R and GLUT-4 of infiltration more than musucularis,lymph node metastasis,III and IV stage of TNM in CC was higher than infiltration not exceed musucularis,without lymph node metastasis,I and II stage of TNM in CC(P<0.05,P<0.05,P<0.05).The protein expression of IGFBP-3 of lymph node metastasis,III and IV stage of TNM in CC was lower than without lymph node metastasis,I and II stage of TNM in CC(P<0.05,P<0.05).The expression of IGFBP-3 was not significantly correlated with depth of invasion in CC(P>0.05).2 The results of Western Blot2.1 The expression of clinic pathological indicators and the expression of IGF-1,IGF-1R,IGFBP-3,GLUT-4 mRNA in CCThe expression of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 mRNA has no connection with age,sex and differentiation in CC(P>0.05).The relative quantity of IGF-1,IGF-1R and GLUT-4 mRNA of infiltration more than musucularis,lymph node metastasis,III and IV stage of TNM in CC was higher than infiltration not exceed without lymph node metastasis,I and II stage of TNM in CC(P<0.05,P<0.05).The expression of IGFBP-3 mRNA was not significantly correlated with depth of invasion in CC(P>0.05).4 Correlation of IGF-1,IGF-1R,IGFBP-3 and GLUT-4 in CCThe results of immunohistochemical,Western Blotting,QRealTime-PCR were consistent: IGF-1 was positively correlated with IGF-1R(r=0.412,P<0.01;r=0.573,P<0.01;r=0.536,P<0.01);IGF-1 was negatively correlated with IGFBP-3(r=-0.604,P<0.01;r=-0.412,P<0.01;r=-0513,P<0.01);IGF-1 was positively correlated with GLUT-4(r=0.396,P<0.01;r=0.568,P<0.01;r=0.658,P<0.01).Conclusion:1 The expression of IGF-1,IGF-1R and GLUT-4 in CC was obviously higher than those in Ad and NM,which shows that three factors may be involved in the intestinal mucosa of canceration process.The expression levels of IGFBP-3 in CC was obviously lower than those in Ad and NM,which prompts that the overexpression of IGFBP-3 may have inhibitory effect on the development of CC.2 The expression of IGF-1,IGF-1R and GLUT-4 of lymph node metastasis,III and IV stage of TNM in CC was higher than without lymph node metastasis,I and II stage of TNM in CC,suggesting that IGF-1?IGF-1R?GLUT-4 play a role in promoting the Colorectal cancer's invasion and metastasis.The expression of IGFBP-3 of lymph node metastasis,III and IV stage of TNM in CC was lower than no lymph node metastasis,I and II stage of TNM in CC,suggesting that overexpression of IGFBP-3 may has inhibitory effect on invasion and metastasis of colorectal cancer.3 In colorectal cancer,IGF-1 was positively correlated with IGF-1R and GLUT-4,IGF-1 was negatively correlated with the IGFBP-3,suggesting that IGF-1R,IGFBP-3,GLUT-4 may through interaction with IGF-1 participates in the development of colorectal cancer.
