| Object: To explore the mi RNA expression profile of Kazakh esophageal squamous cell carcinoma in Xinjiang,and identify the differentially expressed miRNA by using microarray and bioinformatics techniques,the biological function was analyzed after that.in order to further understand the mechanism of the occurrence and development of esophageal squamous cell carcinoma in Xinjiang,and find the new specific molecular markers for the diagnosis and treatment of esophageal squamous cell carcinoma.Methods: Three pairs of ESCC tissues and their adjacent normal esophageal tissues in Xinjiang Kazakh patients were selected and miRNA microarray was used to detect the expression of miRNAs.The differential expression profiles of miRNA was established by means of scatter plot and cluster analysis.Bioinformatics methods were used to predict target genes in differentially expressed miRNA.KOBAS analysis and GO analysis were used to predict the biological functions of differentially expressed miRNA.Results: A total of 23 miRNAs were differently expressed in Kazakh patients with ESCC,with 12up-regulated and 11 were down-regulated.A total of 224 target genes were predicted by mi RWalk.The enriched KEGG pathway of up-regulated miRNAs included Glycosphingolipid biosynthesis-ganglio series,Endocrine and other factor-regulated calcium reabsorption,Steroid biosynthesis.Down-regulated miRNAs were involved in Endocrine and other factor-regulated calcium reabsorption,Steroid biosynthesis,Endocytosis.The GO significant enrichment analysis showed that the up-regulated miRNAs were mainly involved in vesicles,membrane vesicles,the establishment of vesicles,and the biological functions of the exocrine.and down-regulated miRNAs were involved in negative regulation of integrin mediated signaling pathway,lipid binding and so on.Further analysis of the two level entry frequency discovery membrane,organelles,single biological processes,development process,biological regulation,location may be related to the up-regulated miRNA.Membrane,organelles,development processes,positive regulation of biological processes,and multicellular biological processes may be associated with down-regulation of miRNA.Conclusions: there are a large number of differentially expressed miRNA in the ESCC of Kazakh in Xinjiang.Bioinformatics analysis of the screened differentially expressed miRNA obtained a lot of target genes and biological function,The dysregulation of these biological function may relate to the development of ESCC of Kazakh in Xinjiang. |
