Objective:In this research,our purpose was to observe the expression changes of SIRT2,NLRP3 and IL-1 beta in atherosclerotic plaque of Wistar rats,including the level of RNA and protein.And we investigated whether SIRT2 plays an impor-tant role through NLRP3/IL-1 beta signaling pathway.Method:1.Establishment of animal model:The model of atherosclerosis was established 18 old male Wistar rats of 6-week-old feeding with vitamin D3 combined with high fat diet.The basic feeding was given in normal control group.2.Wistar rats was sampled after 2,5 and 8 weeks after high fat diet,we took blo-od serum lipid.The aortic root was used to make paraffin sections and HE staining was used to observe the morphology of the aorta.3.The part of the aortic tissue was used for total RNA extraction to detect the ex-pression of SIRT2,NLRP3 and IL-1 beta RNA levels by PCR.And the protein to as-say the expression of NLRP3 and SIRT2 by Western blotting test.Result:1.Compared with the normal group,the blood lipid level of the model group incr-eased significantly.2.The comparison between the two different groups of Wistar rats at the same ti-me,the mRNA expression level of SIRT2 of Wistar rats in model group was lower than that in normal group,and that decreased with the formation of atherosclerosis.The RNA expression levels of NLRP3 and IL-1 beta in model group were significantly higher than those of normal control group(P<0.05),and the express-ion of Wistar rats was increased with the forming process of atherosclerosis.3.The protein level of NLRP3 in the model group was gradually increased with the prolongation of the formation of atherosclerosis(P<0.05).However,the protein expression of SIRT2 was not detected in the Western blot test.Conclusion:In the forming process of atherosclerosis in Wistar rats,SIRT2 expression at mRNA level decreased accompainied by atherosclerotic plaque formation,while N-LRP3 and IL-1 beta increased.At the protein level by testing,SIRT2 expression was not obvious,but the expression level of NLRP3 increased with the formation of ath-erosclerotic plaque.
