| Objective:To investigate the immune mechanism in experimental autoimmune myasthenia gravis(EAMG)rats treated by the method of invigorating qi for ascending.Methods:48 Lewis rats were randomly divided into 6 groups:positive control group,invigorating qi for ascending astragalus low dose group,invigorating qi for ascending astragalus middle dose group,invigorating qi for ascending astragalus high dose group,CFA group and EAMG model group.The rats were immunized with Ra97-116.Evaluate rats’ weight changes and clinical symptoms and the decrement percentage of RNS.AchR-Ab and IL-10 levels was detected by ELISA in serum.WB was adopted to determine the expression of CD 19 and CD4 protein in thymus tissues.RT-PCR was adopted to determine the expression il-10 and CD 19 and CD4 and FOXP3 genes in thymus tissues.Results:After medication,the percentage of decrement of RNS of rats in invigorating qi for ascending and positive control groups were decreased significantly(P<0.05),and rats’weight and other symptoms were improved significantly(p<0.001);the titers of AChR-Ab was decreased significantly(P<0.05),with the titer of IL-10 increased significantly(P<0.01);The expression of CD 19 protein was down-regulated(P<0.01).The expression of CD4 protein and gene was up-regulated.The expression of CD 19 and IL-10 gene was down-regulated(P<0.O1).The expression of FOXP3 gene was up-regulated(P<0.01).Conclusion:Invigorating qi for ascending groups can mprove body weight and symptoms of EAMG rats.The immune mechanism of treatment may be promot IL-10 content and down-regulate the expression of CD 19 protein and CD 19 and IL-10 gene and down-regulate the expression of CD4 protein and gene and foxp3 gene inhibiting the synthesis of AchR-ab so that the damage of AchR of NMJ will be reduced. |
