Objective:Study on the effect of Flos Trolli total flavonoids on repeated Cerebral Ischemia reperfusion Model in mice,and Rats MCAO Model established by suture method,Effects of oxidative injury and inflammatory cascade reaction and energy metabolism,The main research route of rat MCAO model was to study the effect and mechanism of Flos Trolli total flavonoids on cerebral ischemia reperfusion injury.Methods:After clipped bilateral CCA 15min by microartery clamp,removed bilateral artery clamp,resuscitation of bilateral CCA for 15 minutes,after clamping 15 min,removed the arteriole clamp to recover the perfusion to established repeated cerebral ischemia reperfusion mice model.In Sham group,only separated bilateral CCA,and not made clipping.Sham and MCAO group was given the same volume 0.5%CMC solution.In Nimodipine group,the Flos Trolli total flavonoids were given continuously for 7 days in the large,medium and small dosage groups.1 times/day,making model after administration 1 hour on the 7th morning,ischemia-reperfusion after 22 hours,determined the activity of NSE and the content of S100?in serum by brain sampling,and the level of NO,NOS,ICAM-1,TGF-?1,TNF-?,IL-1?in the brain homogenate,observed the degree of brain injury by HE staining.Protective effect of Flos Trolli total flavonoids on oxidative damage of MCAO Rats Model:Established MCAO Rats Model by Thread method.Use a thread bolt from the left CCA via ICA to enter the middle cerebral artery?MCA?,unplugging after 2 h to achieve reperfusion to establish MCAO rats model.Sham group exposed only left CCA,No thread.Given the corresponding volume of 0.5%CMC solution to Sham and MCAO groups,In Nimodipine group,the Flos Trolli total flavonoids were given continuously for 5 days in the large,medium and small dosage groups.1times/day,making MCAO after administration 1 hour on the 5th morning.ischemia-reperfusion after 22 hours,objective evaluated NDS in Rats,determined the activity of NSE and MPO,and the level of GSH-PX,CAT,MDA,ROS,Bcl-2,Casp-3in brain homogenate,observed the degree of brain injury by HE staining..The same experiment 2 of Flos Trolli total flavonoids on MCAO Rats Model and the way of Drug Administration,ischemia-reperfusion after 22 hours,determined the level of TNF-?,IL-1?,IL-6,NF-?B,IL-10,ASP,GLU,VCAM-1,ICAM-1,ATPase,Coomassie brilliant blue.Study on the effect of Flos Trolli total flavonoids on inflammatory response and energy metabolism in MCAO rats model.Results:In MCAO group,observed the symptoms of mental depression,delayed action and inactive,showed that successfully established the repeated cerebral ischemia and reperfusion model.Compared with MCAO group,FTTF dose group significantly decreased NSE activity and S-100?level in serum,decreased the level of ICAM-1 and the activity of NOS in brain homogenate,and Increase the level of TGF-?1 and NO,significantly alleviated and improved the pathological injury of cerebral cortex.NDS evaluation report shows the success of MCAO rats model.Compared with MCAO group,FTTF dose groups significantly decreased NDS scores,.significant decreased the activity of NSE and MPO in serum,Reduced MDA and ROS content in brain homogenate,Significantly enhanced GSH-PX,CAT activity,Promoting the expression of anti-apoptotic factor Bcl-2,Inhibited of the increase of Caspase-3content,Effective improved pathological injury in the cortical and hippocampal regions of the brain.NDS evaluation report shows the success of MCAO rats model.Compared with MCAO group,FTTF dose group significantly reduced inflammatory factor TNF-?,IL-1?,IL-6,NF-?B,VCAM-1,ICAM-1 induced rat brain homogenates and the level of ASP and GLU.Increasing the content of anti-inflammatory factor IL-10 effectively,significantly enhanced the activity of ATPase.Conclusion:According to the intervention results of Flos Trolli total flavonoids total flavonoids on the repeated cerebral ischemia reperfusion mice model and the MCAO rats model established by the method of thread embolization,the results suggest that:Flos Trolli total flavonoids can effectively inhibit the malignant development of cascade of inflammatory signals in waterfall.significantly improved the symptoms of nerve loss after MCAO.Inhibited the expression of TNF-?,IL-1?,IL-6,Promoting the release of anti-inflammatory factor IL-10 TGF-?1,Significant reducd the level of adhesion molecules,Inhibited peroxidation,Reduced the release of oxygen free radicals,Regulated ROS metabolism,Enhance the activity of antioxidant enzymes,Reducing apoptosis,To effectively control the transcription and expression of the upstream inflammatory signal NF-?B,significantly inhibited the excessive secretion of EAA in and out of cells,enhance the EAA reuptake of the intercellular space,reduce the membrane peroxidation of mitochondria and Ca2+overload,enhance the regulation and supply of ATP in the brain,Improve cerebral oxygen metabolism,Protecting the brain from ischemia-reperfusion damage.
