Objective: To elaucidate the regulation of the inflammatory protein ? from viral macrophages(Virus macrophage inflammatory protein-?,vMIP-?)N-terminal 21 amino acid peptides(NT21MP)in the miRNA expression profile of breast cancer cells.Methods: The MCF-7 cells of breast cancer were grouped into three groups: blank group(B),SDF-1? Group(S)and SDF-1?+NT21MP group(N).When cells grow to70%-80% density,PBS,SDF-1 and NT21 MP are added to each group at 4 degrees,30 min,and N group NT21 MP is added,after 30 min,then added SDF-1? stimulation,placed in CO2 constant temperature incubator.Each groups of cells was collected to extract RNA,and the RNA quality was evaluated and then transcribed into cDNA.The experimental was designed according to the 2-??Ct analytical method,which was usedin the quantitative analysis of SYBR Green I.Testing the difference of miRNA expression profiles between each group.Results: A total of 168 miRNAs genes were detected in order to increase the twice-fold or cut down to 0.5 times-fold screening.Compared with group SDF-1?,there are nine higher in NT21 MP groups: miR-223-3p,miR-451 a,miR-128,miR-489,miR-141-3p,miR-320 a,miR-155-3p,miR-335-3p,miR-320 e,and two lower: miR-15a-5p,miR-339-5p.Conclusion: The differential expression profiles of miRNA were screened by NT21 MP,which may be related to the effect of NT21 MP on breast cancer.
