Wu Shengyuan’s Rule Of Differentiation And Treatment Of GA And The Mechanism Of Qingretongluo Recipe In Intervention Of AGA In Rats

Objective: 1.Combining modern data mining methods,explore Professor Wu Shengyuan’s rule of treating gout medication,with a view to the inheritance and innovation of Professor Wu Shengyuan’s clinical experience in treating gout,and provide reference for clinical treatment of gout.2.To investigate the regulation effect of Qingretongluo recipe on TLRs/ NFκB pathway related factors in the rat model of acute gouty arthritis,and to reveal part of its anti-gout mechanism.Methods: 1.Apply the TCM heritage supporting platform system(V2.5)to the other party and the medical data to carry out statistical analysis and induction,and summarize the drug use rules.2.90 SD rats were randomly divided into 6 groups: normal group,model group,colchicine group,Qingre Tongluo Recipe low,medium and high dose group.Except the normal group,the AGA model was established by injection of MSU solution ankle joint(Modified Coderre method)[1],normal group,model group,daily saline 1ml/kg/d once a day;colchicine group colchicine 9mg/kg/d with distilled water to make suspension The rats in the low,medium and high doses of Qingretongluo Recipe were1.91 g/kg/d and 3.82 g/kg/d,respectively.7.64g/kg/d The extract is diluted into a suspension and administered to the stomach.The rats in each group were given 10 ml/kg/d,once a day for 7consecutive days.Seven days after the intervention,rat peripheral blood mononuclear cells(PBMCs)and plasma samples were taken.RT-PCR,Western-blot,ELISA and other detection techniques were used to observe the TLR-4 and NFκB-P50 of PBMCs in Qingretongluo Recipe.The effects of NFκB-P65,IL-1β m RNA and protein expression,plasma IL-1β,IL-37 expression and their differences,and the effects of Qingretongluo Recipe on TLRs-NFκB and its signaling pathways and their main targets.Results: 1.Through the analysis of 105 prescriptions,involving 79 flavors of raditional Chinese medicine,34 flavors of drugs with frequency ≥10 times,42 classic ombination patterns,6 groups of high-frequency drug associations,51 pairs of commonly used drugs,and evolution 3 The first prescription for the treatment of gout,the drug group is mainly based on hurricane dehumidification and collaterals.2.Qingre Tongluo Recipe can improve the swelling of the ankle joint of rats,compared with the model group(P<0.05).In addition to the normal group,the swelling index reached a peak at 24 h after modeling at each time point in the group,starting at24-72 h.Decreasing gradually,48 h decreased significantly;3.Qingre Tongluo Recipe can reduce the levels of IL-1β and IL-37 in TLR-4,NFκB-P50,NFκB-P65,IL-1β m RNA,protein and plasma in PBMCs(P< 0.05),significantly better than the colchicine group,the results had statistically significant difference(P<0.05).Conclusion: 1.Professor Wu Shengyuan treats gout with medicinal herbs,mostly with clearing heat and collaterals,spleen and dehumidification legislation,observing the pathogenesis,syndrome differentiation and treatment,Xinkai bitterness,cold and heat sharing,Tongbu combination,paying attention to protecting the spleen and stomach,strengthening the righteousness and evil spirits.Formed an unique diagnosis and treatment practice experience,meaning the strong reference significance.2.For Treatment of acute gouty arthritis qingretongluo is an effective prescription,which can down-regulate the expression of TLR-4,NFκB-P50,NFκB-P65,IL-1β m RNA,protein and pro-inflammatory factor IL-1β.The anti-inflammatory cytokine IL-37 is up-regulated and may mediate this signaling pathway as one of its mechanisms ofaction.
