| Objective:Triple negative breast cancer(triple-negative breast cancer,TNBC)is a special type of breast cancer,accounting for 10% and 20% of all breast cancer.Estrogen receptor(estrogen receptor,er),progesterone receptor(progesterone receptor,pr)and human epidermal growth factor receptor-2(human epidermal growth factor receptor-2,her2(EGFR-2)are all negative in breast cancer.Compared with conventional breast cancer,the recurrence and metastasis rate and mortality rate are higher and prognosis is worse,so far there is no effective treatment.β-elemene is an active monomer extracted from traditional Chinese medicine Wen Yu Jin,which has broad-spectrum anti-tumor effect.Its anti-tumor mechanism is mainly to inhibit the migration,invasion and cell cycle of tumor cells,and the mechanism of β-elemene is to inhibit the migration,invasion and cell cycle of tumor cells.Apoptosis of tumor cells was induced,but the effect of β-elemene on protein phosphorylation was rarely studied.Therefore,in order to investigate whetherβ-elemene through protein phosphorylation inhibits the proliferation of triple-negative breast cancer cells,We studied the effect of elemene on apoptosis,proliferation and dry globulation ability of MDA-MB-231 cell line,and discussed the effect of elemene on pi3k/akt,erk/p38 mapk,of breast cancer cell line,and observed the effect of elemene on apoptosis,proliferation and dry globulation ability of triple negative breast cancer cell line.The role of nf-κ b and other key protein phosphorylation levels in the signal transduction pathway,to study the role of elemene in the phosphorylation of breast cancer protein and the effect of elemene on the phosphorylation of breast cancer protein.Methods:(1)The inhibitory effect of β-elemene on breast cancer cell line MDA-MB-231 was significantly inhibited by MTT assay,and detect the correlation between inhibition efficiency and drug concentration.(2)Cloning assay to verify the inhibitory effect of elemene on single breast cancer cells(3)flow cytometry(FCM)was used to detect the apoptosis-promoting effect of elemene on breast cancer cells.(4)Sphere assay to demonstrate the stemness inhibitory effect of elemene on TNBC cells.(5)Use the technology of Western blot was to detect the regulatory effects of elemene on MDA-MB-231 cell line in several key proteins of AKT/P-AKT、PI3K/P-PI3K、ERK/P-ERK、P38/P-P38 MAPK and P65/P-P65 signal pathway.Results:(1)The inhibitory effect of β-elemene on the viability and proliferation of MDA-MB-231 celline was enhanced with the increase of drug concentration.The IC50 of β-elemene was calculated to be 110.13 μg/ ml.(2)Cloning assay confirmed the inhibitory effect of β-elemene on single cell line of triple negative breast cancer.(3)β-elemene could induce apoptosis of MDA-MB-231 cells to a certain extent,and the apoptosis rate was positively correlated with drug concentration.(4)The sphere assay found that the stemness ability of mda-mb-231 cells was also inhibited by elemene,and the volume and number of cells were decreased after drug treatment.(5)β-elemene inhibited the signal pathway of PI3K/AKT,ERK/P38 MAPK,NF-κB in triple negative breast cancer cells,and regulated the activation expression level of its key protein P-AKT,P-ERK,P-P38 MAPK,P65,which decreased with the increase of drug concentration.Conclusion:β-elemene can inhibit the proliferation of mda-mb-231 cells and accelerate the apoptosis of three negative breast cancer cells.Inhibit the function of cell stem cell characteristics,and this function is likely to be achieved by down-regulating the phosphorylation level of tumor cell-associated proteins. |
