Study On The Protective Effect Of Coriolus Versicolor-schisandra Chinensis Composite Extract On Liver Injury In Mice

Excessive alcohol consumption can cause the following damage to human body:alcoholic fatty liver and hepatitis,which can lead to liver fibrosis and even liver failure in severe cases.When the body ingests alcohol,metabolic system is disordered,which leads to oxidative stress,lipid peroxidation in the liver and fatty liver formation.As a classical model of chemical liver injury,CCl₄ liver injury model has similar characteristics with human viral hepatitis.Coriolus versicolor is included in the 2015 edition of the“Chinese Pharmacopoeia”.It has the functions of invigorating the spleen,eliminating dampness,clearing away heat and detoxifying toxins.Modern pharmacological studies have shown that Coriolus versicolor has the functions of protecting liver,analgesia and anti-cancer.Schisandra chinensis was first recorded in“Shennong Herbal Classic”and listed as the top grade.Modern pharmacological studies have shown that Schisandra chinensis has many active ingredients and good pharmacological effects on many diseases.It can be used in combination with other traditional Chinese medicines to treat and alleviate acute and chronic liver injury.On the basis of the hepatoprotective effect of Coriolus versicolor and Schisandra chinensis extracts,the compound Chinese herbal medicine of them was prepared for the first time in this paper,and its hepatoprotective effect was studied.The main results of this study are as follows:1.Study on the protective effect of Coriolus Versicolor-Schisandra Chinensis composite extract on alcoholic-induced liver injury in mice.Liver pathological changes were observed by organ index,H&E staining,serum liver function index and liver tissue antioxidant index.To study the protective effects of Coriolus versicolor-Schisandra chinensis（CS）on acute alcoholic liver injury in mice.The results showed that CS could significantly inhibit the increase of liver index and the decrease of spleen index caused by alcohol;and improve the exfoliation and steatosis of liver cells and oxidative stress.Furthermore,it could also significantly reduce the levels of ALT,AST,MDA,TG,TC and LDL-C;and increase the content of HDL-C in serum.Moreover,it could also improve the activities of SOD,GSH-Px and CAT in liver tissue.This suggested that CS composite extract have a good hepatoprotective effect on alcoholic liver injury.2.Study on the protective effect of Coriolus Versicolor-Schisandra Chinensis composite extract on CCl₄-induced liver injury in mice.Liver pathological changes were observed by organ index,H&E staining,serum liver function index,inflammatory factors and antioxidant index of liver tissue.To study the protective effects of CS on CCl₄ liver injury in mice.The results showed that CS could significantly inhibit the increase of liver index and decrease of spleen index caused by CCl₄,improve the fragmentation necrosis and steatosis of liver cells,significantly reduce the levels of serum ALT,AST,TBIL,TG,TC,LDL-C,TNF-α,IL-6 and IL-1β,and increase the level of HDL-C;Moreover,they could also significantly reduce the level of MDA in liver tissue and increase the levels of SOD,GSH-Px,CAT activities.These results suggested that CS composite extract have a good hepatoprotective effect on CCl₄-induced liver injury.
