Study Of The WULING Tablet As Health Food For Reducing Alcoholic Liver Injury

WULING Tablet is a health food composed of Schisandra chinensis and Ganoderma lucidum powder to reduce the risk of alcoholic liver injury.Alcoholic liver injury is a disease that causes severe liver damage and liver cell structure damage due to prolonged drinking.The degree of injury was related to alcohol intake and time of alcohol intake.According to the different degree of injury can be divided into alcoholic fatty liver,alcoholic hepatitis,liver fibrosis and liver cirrhosis four stages.Objective:To study the extraction technology of Schisandra chinensis and Ganoderma lucidum in as a health food for alcoholic liver injury provides a theoretical basisMethods:1.The technology of WULING Tablet was studied: the extraction process of schisandra,the preparation of Ganoderma lucidum powder and the investigation of technological conditions;2.Drafting of quality standards(draft): A preliminary study on the stability of WULING Tablet,including prescriptions,methods,content determination and WULING Tablet;3.Study on protective effects of alcoholic liver injury:Male Wistar rats,a total of 200,weight 180~200 g,randomly divided into two groups,12 only for the normal group,gavage at 0.5 % carboxymethyl cellulose sodium,the remaining 188 for model group,1th to 4th week gavage at 40 % Liquor 8 g/kg,5th to 8th week gavage at 50 % liquor 9g/kg,The 9th to 16 th week gavage at 50 % liquor 10 g/kg.The 9th Week model group was randomly divided into 8 groups according to weight,namely: The Model control group was given 0.5 % carboxymethyl cellulose sodium,WULING Tablet prescription 0.25,0.5,1.0 g/kg dosage Group,WULING Tablet prescription two 0.25,0.5,1.0 g/kg dosage Group,positive control group to give water silymarin 80mg/kg,each group of the stomach for medicine,1 times daily.8 weeks after the drug delivery,10 % Chloral hydrate 30mg/kg,intraperitoneal injection of anesthesia rats,abdominal aorta blood intake,liver,test indicators are as follows:Serum: alanine aminotransferase(ALT),aspartic acid aminotransferase(AST),triglyceride(TG),total cholesterol(TC),very low density lipoprotein(VLDL),free fatty acid(FFA),Malondialdehyde(MDA),superoxide dismutase(SOD);Liver tissue: triglyceride(TG),total cholesterol(TC),Malondialdehyde(MDA),superoxide dismutase(SOD),liver index and liver pathological changes.Results:1.The best preparation process of the WULING Tablet:Prescription one: flavor of medicinal herbs-ganoderma lucidum medicinal herbs =1:1.Schisandra chinensis Extract hydroxyethyl beta cyclodextrin cyclodextrin =1:2,Ganoderma lucidum-starch =1:2,adhesive for 10%PVP,wetting agent 50% ethanol solution.Prescription two:flavor of medicinal herbs-ganoderma lucidum medicinal herbs =2:1.Schisandra chinensis Extract hydroxyethyl beta cyclodextrin cyclodextrin =1:2,Ganoderma lucidum-starch =1:2,adhesive for 10%PVP,wetting agent 50% ethanol solution.2.The initial stability of the WULING Tablet is better;the quality standard(draft)of the WULING Tablet was drafted and can be used for quality control of WULING Tablet.3.Efficacy research of WULING Tablet:(1)Compared with normal group,the content of TC,TG,VLDL and FFA in model group was obviously elevated,the content of TC and TG increased(p<0.05;p<0.001),compared with the model group,the 1.0g/kg group and Silymarin group could decrease the TC of rat Serum and liver tissue significantly,TG content and serum VLDL,FFA content(p<0.05),WULING Tablet two,0.5,1.0 g/kg group can significantly reduce the rat serum and liver tissue TC,TG content and serum VLDL,FFA content(p<0.05 or p<0.01),in the same dosage,WULING Tablet two group is superior to WULING Tablet on first group;(2)Compared with normal group,the activity of serum AST and ALT in model group was significantly elevated(p<0.05 or p<0.01),compared with the model group,the 1.0 g/kg group of WULING Tablet could significantly reduce the serum alt activity(p<0.05),WULING Tablet two,0.5,1.0g/kg group and the Silymarin group could significantly reduce the serum ALT activity(p<0.05 or p<0.01)in rats,and the effect of the WULING Tablet two was better than that of the WULING Tablet one.All groups had no significant effect on serum AST activity in rats;(3)Compared with normal group,the content of MDA in serum and liver tissue of model group was obviously elevated,SOD activity decreased(p<0.05 or p<0.01),compared with model group,WULING Tablet 1.0 g/kg Group,WULING Tablet two,0.5,1.0 g/kg group and Silymarin group could significantly reduce the content of MDA in serum and liver tissue of rats,elevate SOD activity(p<0.05 or p<0.01),and the effect of WULING Tablet two was better than that of WULING Tablet one.(4)Compared with the normal group,in the model group,the weight of rats was significantly decreased,the liver index was significantly elevated(p<0.01),compared with the model group,the body weight and liver index of the WULING Tablet in each dosage group and the Silymarin group had no significant improvement.(5)Compared with the control group,the hepatic cells in the model group were seen as circular vacuoles,the nucleus was elliptical,and the hepatic sinus was enlarged.Compared with the model group,the pathological changes of hepatocytes were reduced in the prescriptions of WULING Tablet and the two prescriptions,and the prescription two and prescription were no difference.Conclusion:1.Two kinds of preparation technology of WULING Tables(schisandra medicinal herbs-ganoderma lucidum medicinal herbs =1:1;Schisandra chinensis Medicinal herbs-ganoderma lucidum medicinal herbs =2:1)can be Feasible.2.The two preparation techniques of WULING Tablet are of controllable quality and good initial stability.3.Efficacy screening of WULING Tablet:(1)WULING Tablet can significantly reduce serum AST and ALT vigor and improve liver function.2.WULING Tablet can significantly reduce the level of lipid deposition in rat serum and liver tissue and regulate lipid metabolism.3.WULING Tablet can significantly reduce the content of MDA in serum and liver tissue,and significantly increase sod activity.WULING Tablet can reduce lipid peroxidation,enhance the body antioxidant ability,reduce liver cell lipid peroxidation injury.Reduce the degree of liver cell necrosis.4.The above effect of prescription two is better than prescription one,and prescription two is used as a guarantee to reduce the harm of alcoholic liver injury Health food,to be developed and used.
