| Objects:Colorectal cancer includes rectal and colon cancer.Colon cancer is the third most common cancer in the world.However,20 to 30 percent of colorectal cancer patients have a family history of colorectal cancer,and 5 percent of these tumors are consistent with Mendelian genetic law.In many patients,there are benign tumor lesions:adenomatous polyps or filamentous polyps before colorectal cancer.Studies conducted over the past few years have described major molecular changes in colorectal cancer,showing that each patient’s tumor has undergone multiple molecular changes,including gene expression.Early detection of differentially expressed proteins in colorectal cancer is helpful for early treatment before malignant tumors,which will greatly improve the prognosis and survival rate of patients.VIM is overexpressed in various cancers and is closely related to the occurrence and development of tumors,and is a potential therapeutic target for cancer.In order to further understand the correlation between VIM and colorectal cancer,The differential expression of proteins in human colorectal cancer tissues and healthy tissues were detected and analyzed by immunoblotting.The lentiviral vector was used to interfere with the expression of VIM,and its effect on the biological activity of tumor cells was observed.It is expected to provide help for the treatment and prognosis of colorectal cancer.Method:1.Fresh tumor specimens of colorectal cancer from December 2017 to December 2018 were collected from the first affiliated hospital of jilin university as the experimental group,and fresh specimens of normal tissues 10 cm away from the pathological tissues were taken as the normal control group.Immunoblot was used to detect the protein expression in colorectal cancer specimens and the control group.2.The lentiviral expression vector was constructed to observe the effect of RNA interference VIM gene expression on the biological function of colorectal cancer cell line SW480,so as to further understand the biological function of VIM.Results:1.Immunoblotting experiments confirmed the up-regulated expression of protein VIM in colorectal cancer tissues.2.The lentiviral vector that interferes with VIM expression was successfully constructed,and the colorectal cancer cell SW480 was successfully transfected to obtain a cell line that interfered with the expression of VIM protein.3.Observing the SW480 cells after transfection,the cell proliferation was significantly inhibited.Conclusion:VIM expression of differentially expressed protein in colorectal cancer tissues is relatively higher than that in adjacent tissues.The differential expression of VIM may be related to the occurrence,development,invasion and metastasis of colorectal cancer,which is a promoting factor for the occurrence and development of human colon cancer and a potential indicator for diagnosis,treatment and prognosis of colon cancer.However,other specific mechanisms in the progression of colorectal cancer need further study. |
