| Objective:Adopt a network pharmacology research method,Obtaining the"ingredient-target"network of ulcerative colitis with Huangqixiangshen Decoction,and predicting the biological pathway nature of Huangqixiangshen Decoction in the treatment of ulcerative colitis,and providing a basis for the scientific nature of traditional Chinese medicine theory.Methods:Using the ADME parameters of pharmacokinetics as the screening criteria,the active constituents of Astragalus membranaceus Maxim,were screened in the TCMSP database platform,and the structures of these active ingredients were submitted to PharmMapper to obtain potential targets for each active ingredient.A database of ulcerative colitis-related targets was constructed using a web database platform(TTD,DrugBank,DisGeNET,OMIM,STRING).After finishing the above targets by Excel 2016,the predicted target of UC in the treatment of UC was obtained,and Cytoscape was used to construct the predictive target network of ulcerative colitis with Huangqixiangshen Decoction,and the target was predicted by The Gene Ontology(GO).The bioprocess analysis was performed at the point,and the bio-enrichment of the predicted target was performed by Reactome FI,and the possible action pathway of Huangqixiangshen Decoction in treating UC was obtained.RESULTS:Oral availability(OB%)≥30%,drug-like(DL)≥0.18 as parameters,screening 189 Huanghuaxiangshen soup active ingredients.A total of 845 ulcerative colitis-related targets were obtained by TTD,DrugBank,DisGeNET,OMIM,and STRING.After finishing,14 Baiji therapeutic UC target were predicted,Diyu was 20,Huangqi was35,Kushen was 37,Muxiang was 14,Sanqi was 14,Gancao was 31,and the predicted target of cadmium for the treatment of UC,finally obtained the predicted target of 44 jaundice ginseng decoction in the treatment of UC.GO bioprocess analysis and Reactome FI biochannel enrichment screening for predicted targets with P value<0.001,44 predicted targets involved in cellular metabolic processes,protein metabolism,signal transduction,immune system,and regulation of organism response to stress The biological process involves 27 signaling pathways,which are mainly Toll-like receptor pathway,NLR receptor pathway,NOTCH signaling pathway,TNF signaling pathway,and TP53 gene regulatory pathway.Conclusion:Huangqixiangshen Decoction may pass the Toll-like receptor pathway(target APP,BTRC,MAP2K6,MAPKAPK2,TAB1),NLR receptor pathway(target APP,MAP2K6,CYLD,TAB1)and TNF signaling pathway(target RNF31,CYLD),TAB1)inhibits NF-κB activation and release of inflammatory factors downstream of MAPK signaling,promotes intestinal barrier recovery by inhibiting overactivation of Notch signaling pathway(targets CCND1,EP300,KAT2B),and regulates TP53 by acetylation(target EP300,MAP2K6)contributing to prevention and treatment of UC intestinal mucosal malignant transformation.This indicates that Huangqixiangshen Decoction plays a role in the treatment of UC from multiple targets,multiple angles and multiple levels,which provides a direction for the experimental research of Huangqixiangshen Decoction in the future. |
