Discussion On The Mechanism Of Shaoyao Decoction In The Treatment Of Ulcerative Colitis Based On The Auxiliary Platform Of Traditional Chinese Medicine Inheritance And Network Pharmacology

Objective : Using TCM Inheritance Assistant Platform and Internet Pharmacology to Explore the Mechanism of Shaoyao Decoction in Treating Ulcerative Colitis.Methods:The literature comes from January 1949 to December 2020 and is included in the four major databases(China Journal Full-text Database(CNKI),Wanfang Data Knowledge Service Platform,Chongqing Weipu Database,Duxiu System,etc.)on the treatment of ulcerative colon by Shaoyao Decoction The retrieval time of modern medical literature of Yanyan is limited to January 1,1949 to December 31,2020.Those who meet the criteria for inclusion are entered into the TCM inheritance auxiliary platform to analyze the rules of prescription,dosage,decoction,and treatment.Using network pharmacology to predict the effective target and mechanism of Shaoyao Decoction in the treatment of ulcerative colitis.Results:1.Auxiliary platform for Chinese medicine inheritance(1)Document inclusion results: A total of 23,033 documents were retrieved from How Net,4,251 from Wanfang,6,379 from Weipu,3862 from Duxiu,and 117 documents were obtained after two screenings;(2)In the prescription statistics,the spleen meridian,stomach meridian and large intestine meridian are used with the highest frequency;the cold medicine is the highest frequency used for the four qi,followed by warm medicine;the frequency of use of the five flavors is as follows: bitter>Pungent> sweet> sour> astringent> salty;(3)In the drug mode,the most frequently used drug pairs are "Scutellaria Baicalensis & Coptis","Scutellaria Baicalensis & White Peony","Coptis &White Peony","Scutellaria Baicalensis & Muxiang";in the analysis of association rules,"Atractylodes->Coptis" Fangfeng->Coptis","Citrus chinensis peel->White peony","Fangfeng->White peony root","Citrus peel->Muskum" have higher confidence;the top 20 medicines with higher frequency of use are: Scutellaria baicalensis,Coptis chinensis,and White peony root,Woody,angelica,betel nut,cinnamon,rhubarb,licorice,roasted licorice,pulsatilla,sanyu,tangerine peel,parsnip,atractylodes,honeysuckle,citrus aurantium,locust,Bupleurum,fried atractylodes;(4)Based on entropy clustering algorithm,5 potential new party combinations can be evolved;2.Network Pharmacology(1)About Shaoyao Decoction: 149 active ingredients of Shaoyao Decoction,151 effective compound target proteins,151 gene targets,1111 ulcerative colitis disease targets,and 82 common targets were obtained through Biso Genet.The biological processes involved in the target include viral processes,double-strand break repair through non-homologous end joining,etc.;the cell components involved include nucleoplasm,nucleus,etc.;the molecular functions involved include protein binding,poly(A)RNA binding,etc.;The signal pathways involved include viral carcinogenesis and MAPK signal pathways.(2)About Shaoyao Decoction Recipe: 205 active ingredients in Shaoyao Decoction Recipe,159 effective compound target proteins,159 gene targets,and 1111 disease targets for ulcerative colitis were screened out.Commonly obtained through Biso Genet There are 152 targets.The biological processes involved in these targets include translation initiation,SRP-dependent co-translational protein targeting membranes,etc.;the cellular components involved include nucleoplasm,nucleus,etc.;molecular functions involved include protein binding,RNA binding,etc.;The signal pathways involved include viral carcinogenesis and MAPK signal pathways.Conclusion: 1.The statistics of the four qi and five flavors found that the four qi of drugs are mainly cold and warm(cold medicine is slightly higher than warm medicine),and the five flavors of medicine are mainly bitter and pungent(bitter medicine is slightly higher than pungent medicine).The meridians mainly focus on the spleen,stomach,and large intestine meridian.It can be seen that modern physicians use Shaoyao Decoction to treat UC from the spleen,stomach,and large intestine,together with medicines for bitter cold and pungent temperature.2.In the section of drug association rules,it can be seen that the drugs that are strongly related to the original drugs are Atractylodes macrocephala,Fangfeng,Chenpi,Pulsatilla,etc.,which are respectively tonic drugs,anti-epidermic drugs,qi regulating drugs,and heat-clearing drugs.Bitterness and warmth are the mainstays,and the spleen is the main function of the meridian.3.Drug frequency section.The top 20 medicines include white peony,scutellaria,coptis,rhubarb,cinnamon,licorice,betel nut,woody,angelica,pasqueflower,atractylodes,Bupleurum,tangerine peel,scutellaria,parsnip,sophora japonica,honeysuckle,Citrus aurantium,etc.,the composition of the medicine constitutes the Shaoyao Decoction Recipe(that is,the new prescription),which not only has the functions of clearing heat,removing dampness,detoxification,regulating qi,and promoting blood stagnation contained in the Shaoyao Decoction,but also has the functions of soothing the liver The power of strengthening the spleen,cooling blood and stopping dysentery.From the perspective of modern pharmacology,the prescription has multiple effects such as anti-inflammatory,anti-oxidation,and immune regulation.4.Potential new recipe combinations extracted based on entropy algorithm are: Qinpi-Purslane-Sunflower Astragalus-Baiji-Honeysuckle-Agrimonia,Cinnamon-Rhubarb-Pulsatilla-Citrus aurantium-Angelica-Tangerine peel,White peony-White peony-Fried white peony-Baiji-Diyu charcoal,Fructus Aurantii-Cinnamon-Angelica-Betel nut-Tangerine peel-Sophora japonicus,Poria-Panax notoginseng-Cangzhu-Fried Diyu-Coptis-Rooted Astragalus,and its drug compatibility is mostly heat-clearing and detoxifying drugs,Hemostatic drugs,tonic drugs,and Qi regulating drugs,which can treat UC to a certain extent,but the combination of medicines is complicated and irregular,and clinical research is needed.5.In the part of network pharmacology,there are two similarities between Shaoyao Decoction and Shaoyao Decoction(the new prescription):First,the effective compounds of both are mainly flavonoids;both GO-CC,The results of GO-MF and KEGG analysis are roughly the same;the difference is that most of the core targets of Shaoyao Decoction are involved in the occurrence of anti-inflammatory and immune responses;while the core targets of Shaoyao Tang Hua Fang Fang focus on tumor occurrence and development,which may indicate that the formula It may be useful for treating colon cancer or other cancers.6.Shaoyao Decoction and its chemical prescriptions may be achieved by regulating the MAPK pathway in the treatment of UC.
