Objective:Colorectal cancer is a common malignant tumor in clinic.Influenced by dietary habits and other factors,the incidence of colorectal cancer in China has been on the rise in recent years.which poses a major threat to the health of Chinese people.Chemotherapy is a common adjuvant therapy for colorectal cancer.It is widely used in preoperative neoadjuvant therapy to increase the chance of operation,enhance the efficacy after operation to kill residual cancer cells and palliative treatment for advanced patients.However,there are still some problems such as low sensitivity to colorectal cancer cells and obvious side effects.Shenmai injection is a compound preparation composed of red ginseng,Ophiopogon japonicus and so on,which has the effect of Invigorating Qi and nourishing yin.Literature shows that it can enhance the anti-tumor effect of chemotherapeutic drugs when combined with chemotherapeutic drugs in clinic,but the basic research is not perfect at present.The mechanism of Shenmai Inj ection in the treatment of colorectal cancer is not very clear,so it is necessary to study it further.By observing the effect of Shenmai injection combined with oxaliplatin on transplanted colon cancer LoVo cells in nude mice,the mechanism of synergism and attenuation of Shenmai injection combined with oxaliplatin was explored.Methods:The in vivo model of LoVo cell transplantation in nude mice was established.The tumor volume was recorded and the tumors were drawn every 2 days from the day of administration in saline group(n=8),oxaliplatin group(8 mg/kg,n=8),combined treatment group(8 mg/kg,0.01 ml/g,n=8)and Shenmai group(0.01 ml/g,n=8).Tumor growth curve.After 14 days of administration,the effects of oxaliplatin on transplanted tumors were observed.Blood samples were taken from mice and spleen and tumor tissues were taken.The expressions of hypoxia inducible factorla(HIF-la),basic fibroblast growth factor(b-FGF),smooth muscle actin(a-SMA)and platelet-endothelial cell adhesion molecule(CD3 1)were detected by immunohistochemistry.Result:The anti-tumor rate of Shenmai injection combined with oxaliplatin was 49%.The anti-tumor effect of Shenmai injection combined with oxaliplatin was significant and had no obvious toxicity to the blood and liver and kidney of tumor-bearing mice.The combined treatment group could significantly reduce the expression of HIF-1a and b-fibroblast and the microvessel density(MVD)of tumor tissue.Conclusion:Shenmai injection can effectively alleviate the adverse effects of oxaliplatin on immune organs and liver function in nude mice,and enhance its therapeutic effect on colon malignant tumors.Its mechanism may be related to the inhibition of b-fibroblast mediated angiogenesis signaling pathway.
