Objective: In this study,genomic DNA of peripheral blood of children with Kawasaki disease(KD)and healthy control group in Nanchong area was taken as the research object.The nicotinamide adenine dinucleotide phosphate oxidase C242 T locus was analyzed to explore the correlation between the polymorphism of NADPH oxidase p22 phox subunit C242 T gene and the occurrence of Kawasaki disease and its coronary artery lesion,which was the complication of Kawasaki disease and coronary artery lesion.The basis of diagnosis and treatment is provided.Methods: The blood samples of 75 children with Kawasaki disease in Nanchong area from December 2017 to December 2018 were collected.A case-control study was conducted.75 children with KD were selected as the case group and 75 children with outpatient health examination in our hospital as the control group.According to the presence or absence of coronary artery lesion(CAL),the children with KD could be divided into two groups: coronary artery lesion group(CAL)and no coronary artery lesion group(NCAL).Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)combined with direct gene sequencing technology was used to detect the C242 T gene polymorphism of the subjects.The genotype and allele frequencies of C242 T in KD group and control group,CAL group and NCAL group were compared.Results: There was no significant difference in genotype frequency(χ2=0.667,P=0.414)and allele frequency(χ2=0.560,P=0.454)between Kawasaki disease group and healthy control group(P>0.05).The frequency of C242 T genotype(χ2=4.38,P=0.036)and allele frequency(χ2=4.509,P=0.034)of p22 phox subunit were significantly different between CAL group and NCAL group(P<0.05);the frequency of T allele(17%)in NCAL group was significantly lower than that of C allele(83%).CONCLUSION: The C242 T gene polymorphism of p22 phox subunit may not be associated with the susceptibility to Kawasaki disease.The C242 T gene polymorphism of p22 phox subunit may be associated with coronary artery lesion in Kawasaki disease.The C242 T allele T of p22 phox subunit may have protective effect on coronary artery lesion in Kawasaki disease.
