| Objective:To explore the association between loci rs1333049and rs4977574 of CDKN2BAS gene polymorphism with Kawasaki disease(KD)and complicated with coronary artery lesion(CAL).Methods:According to the diagnostic criteria of KD,100 children with KD who were hospitalized in the Third Xiangya Hospital of Central South University were randomly selected in the Kawasaki disease group(KD group),who had not adminstrated aspirin or gamma globulin before admission.There were 25 cases with KD complicated with CAL(KD-CAL)group and 75 cases without CAL(KD-NCAL)group.In addition,a total of 100 healthy children in Hunan Province during the same period were selected as the control group,without medical history of KD,rheumatic,cardiovascular or infectious diseases.In this study,the DNA was extract from peripheral blood samples of two groups and detected the genotype of loci rs1333049 and rs4977574 of CDKN2BAS gene by polymerase chain reaction-direct sequencing(PCR-SBT)technology.The genotype and allele frequencies were calculated.Chi square was employed to test whether the difference between the two groups was significant.Results:For locus rs1333049 in CDKN2BAS gene,there was no significant difference was found between the KD groups and healthy controls in genotyping frequencies of GG,CG,CC and allele frequency of G,C(χ~2=2.448,0.8145;P=0.2941,0.3668).For locus rs4977574 in CDKN2BAS gene,there was no significant difference found between the KD groups and healthy controls group,in genotyping frequencies of AA,AG,GG and allele frequency of G,G(χ~2=3.9885,1.6911;P=0.1361,0.1935).There was no significant difference was found in genotype distribution and allele frequency between the KD-CAL group and KD without CAL(KD-NCAL)group in locus rs1333049 or locus rs4977574of CDKN2BAS gene.Conclusion:The association were not found between loci rs1333049and rs4977574 polymorphism of CDKN2BAS gene and the pathogenesis of KD or KD complicated with CAL. |
