| Objective: Through experiment on living renal material treatment of adriamycin nephrosis rats of albuminuria,observe 24 hours urinary protein quantitative urine trace albumin/uric creatinine and urea nitrogen,serum creatinine and cholesterol,serum albumin,of rats,and renal morphologic indicators changes of podocyte synaptopodin,F-actin,alpha actinin-4expression,provide a theory basis for kidney material treatment of adriamycin nephrosis rats proteinuria.Methods: Using the methods that establishing the rat model of adriamycin nephropathy by single injection of adriamycin into tail vein.The patients were divided into four groups: treatment group(huo shen jiangzhuo fang group),control group(shenyan kangfu tablet group),model group and blank group.The changes of general state,urinary microalbumin/urinary creatinine,24-hour urinary protein,serum creatinine,urea nitrogen,total cholesterol and plasma albumin in rats were observed dynamically.The renal sections were stained with HE to observe the pathological changes of the renal tissue of adriamycin nephropathy rats.The pathological structure of kidney was observed under light microscope.The synaptopodin,f-actin,-actinin-4 expression was observed by immunohistochemical PV two-step method.Results: 1.The treatment group could improve the general status of adriamycin nephropathy rats;2.Serum creatinine,urea nitrogen,total cholesterol,uric microalbumin/uric creatinine and 24-hour uric protein were all lower in the treatment group than in the model group,and plasma albuminin in the model group were higher,the difference was statistically significant(P<0.05).2.The group of huoshenjiangzhuo decoction can alleviate the pathological injury of chronic kidney disease;3.The immunohistochemical results show that compared with the treatment group,control group and blank group,model group synaptopodin,F-actin in glomerular endothelial cells and mesangial cells and epithelial cells express to reduce,the difference was statistically significant(P < 0.05),alpha actinin-4 in glomerular endothelial cells,the expression of mesangial cells and epithelial cells increased,the difference was statistically significant(P <0.05);The decreased expression of a-ctinin-4 at the end of week 8 showed statistically significant difference(P<0.05),while compared with the control group,while the increased expression of synaptopodin and f-actin in the treatment group showed statistically significant difference(P<0.05).Conclusion: 1.Huoshenjiangzhuo decoction can reduce the 24-hour urinary protein quantification and urinary microalbumin/urinary creatinine ratio in adriamycin nephropathy rats;2.Huoshenjiangzhuo decoction can improve serum albumin content and reduce serum cholesterol content in adriamycin nephropathy rats;3.By up-regulating synaptopodin and f-actin protein expressions in renal tissues of adriamycin nephropathy rats and down-regulating alpha-actin-4,the active kidney turbidity reducing prescription may repair multiple damaged podocytes,maintain their intact structures,play their normal functions and reduce urinary protein excretion. |
