| The product specification grade of Chinese herbal medicine is the embodiment of the quality of Chinese medicine,and it is the most intuitive method to evaluate the quality of Chinese herbal medicine.Forsythia is the dried fruit of the Forsythia suspensa(Thunb.)Vahl of the Oleaceae plant.It is one of 40 commonly used Chinese herbal medicines.In the "76 kinds of Chinese herbal medicines product specification grade specification",according to the appearance characteristics and the time of collection,the forsythia medicine is divided into two specifications,"Green Forsythiae Fructus" and "Old Forsythiae Fructus ".However,with the continuous changes in the production and circulation of Chinese herbal medicines and the continuous updating of quality standards,the original standards can no longer meet the current production,circulation and use of forsythia.In this study,literature review,on-site investigation and physical and chemical testing were used to construct the Forsythia commodity specification grade standard based on the identification of sensory properties-chemical quality control-biological titer.The main research contents and results are as follows:A.Division of the standard grade standards of Forsythia medicinal herbs1 Base source identificationUsing the method of literature collation,the research on the basic source of Forsythiae Fructus was carried out.It was found that the use of Forsythia medicinal materials before the Song Dynasty was rather confusing.It was not until the Song Dynasty that the Forsythiae Fructus of Oleaceae.was the only authentic source of Forsythia.Using the method of on-site investigation,the main mixed products circulating in the whole country were investigated.It was found that the main counterfeits of Forsythia in the current market are singular Forsythiae,northeast Forsythiae,and Qin Forsythiae.Finally,traits,microscopic,thin layers and characteristic maps were used to identify forsythia medicinal materials,and the samples were identified as genuine Forsythiae.2 Study on the grade of Forsythia’s commodity specifications based on the main sensory traitsUsing the Delphi method,the main grading indicators of Forsythia were investigated.It was found that the main factors determining the grade of Forsythia were ranked according to their importance: traditional sensory indicators such as color,fullness,gas and taste.This study mainly uses modern instruments to quantify traditional sensory indicators and reduce the subjective factors in the Forsythia grade standard.a)Through the color difference meter to collect the color data of Forsythia,use the collected L,a,b indicators to determine the range of chromaticity values of Green Forsythiae Fructus and Grown Forsythiae Fructus,distinguish the Green Forsythiae Fructus and the old cockroaches by color,and can The old cockroaches are distinguished,but the green medicinal herbs cannot be distinguished from the green cockroaches;b)The odor of 97 batches of Forsythia was collected by electronic nose,and the collected data was modeled by DFA method.The specifications and grades of Forsythia were differentiated according to the interval of the sample.In addition,the medicinal materials and Green Forsythiae Fructus could be distinguished.c)The electronic tongue was used to collect the taste of 97 batches of Forsythia,and the collected data was modeled by DFA method,and the specifications and grades of Forsythia were classified according to the interval of the sample.3 Research on the specification of Forsythia’s commodity specifications based on chemical quality controlAccording to the research results of this subject,with reference to the 2015 edition of the Chinese Pharmacopoeia,the research on Green Forsythiae Fructus and Grown Forsythiae Fructus from different places was carried out from the aspects of inspection and leachate.The main contents are as follows: Impurities,moisture,total ash,residual amounts of heavy metals,pesticide residues,aflatoxins,and sulfur dioxide were measured;alcohol-soluble extracts,water-soluble extracts.It is stipulated that the impurities of Green Forsythiae Fructus should be no less than 3%,the content of alcohol-soluble extract should be no less than 30%,the content of water-soluble extract should be no less than 15%,the content of old-style impurities should be no more than 9%,and the content of alcohol-soluble extract should be no less than 16%,the water-soluble extract content should be no more than 10%.It is stipulated that the moisture of Forsythia medicinal herbs should be no more than 10%,the total ash content should be no more than 4%,the residual amount of heavy metals: lead should be no more than 5.0mg/kg,cadmium should be no more 0.3mg/kg,mercury should be no more than 0.2mg/kg;arsenic should be no more than 2.0mg /kg,copper should be no more than 20.0mg/kg;total 666 should be no more than 0.2mg/kg,total drip should be no more than 0.2mg/kg,pentachloronitrobenzene should be no less than 0.1mg/kg;aflatoxin B1 residue should be no more than 5μg/kg;the total amount of aflatoxin G2,G1,B1,B2 should be no more than 10μg/kg;the residual amount of sulfur dioxide should be no more than 150mg/kg.HPLC method was used to determine the content of Forsythiaside A,and the classification index was determined to be no less than 1.00% selected as model cells,LPS induced RAW264.7 cells to activate them,and an in vitro inflammation model was established.MTT assay was used to detect cytotoxicity.Forsythia extract concentration was in the range of 1.9~16.6μg/mL for mice.RAW264.7 cells have no toxic side effects.The levels of IL-1β and TNF-α inflammatory factors were detected by ELISA.Compared with LPS group,the concentration of Forsythia suspensa extract was significantly anti-inflammatory at 1~5μg/mL.The concentration of five standard samples was 40~480ng/L.In the range,the anti-inflammatory effect on IL-1β inflammatory factors: quercetin> Forsythiaside > rosin-β-D-glucoside> Forsythiaside A> forsythin;Anti-inflammatory effect on TNF-α inflammatory factors: quercetin effect is significantly higher than the anti-inflammatory effect of Forsythiaside,anti-inflammatory effect of Forsythiaside A,forsythin,rosin-β-D-glucoside Not obvious.The anti-viral activity of Forsythia suspense extract was detected by a neuraminidase inhibitory enzyme(NA)inhibitor screening kit.The extract concentration ranged from 0.56 to 135 mg/mL,and the optimal inhibitory concentration was 5 mg/mL.The NA inhibition rate was 61.4%~75.6%,and the NA inhibition rate of the old cockroach extract was 73.0%~99.8%.When the concentration of standard was 0.05~1.00 mg/mL,the NA inhibition rate of Forsythiaside A was the highest,and the antiviral effect of quercetin increased with the increase of concentration.Forsythin,rosin-β-D-glucoside,and Forsythiaside are not associated with antiviral effects.Using SPSS software for significant analysis,the virus inhibition rate of Green Forsythiae Fructus selection was significantly higher than that of Green Forsythiae Fructus currency;the virus inhibition rate of old selection
